桑杏护肺颗粒治疗大鼠放射性肺纤维化的实验研究 被引量:5

摘要 目的:观察桑杏护肺颗粒对放射性肺纤维化模型大鼠的肺保护作用。方法:SD雄性大鼠64只,随机分为空白组、模型组、强的松组(3.125mg/kg)、桑杏护肺颗粒高、中、低剂量组(分别按生药总量20 g/kg/d、10 g/kg/d、5g/kg/d剂量给药)。直线加速器对除空白组外所有大鼠行单次全胸照射,辐射总量14 GY,建立放射性肺损伤模型,造模第二日起按剂量给药。于造模后第4、8、12、16、20、24、28、32w,取大鼠尾静脉血,采用ELISA法检测各组血清炎症因子IL-6、TNF-α改变情况。在造模32w后,处死大鼠收集其肺组织,灌洗后行石蜡包埋、切片。采用HE、Masson染色,并使用肺泡炎评分(Szapiel评分)评价各组肺组织损伤及纤维化程度。结果:与空白组相比,辐射引起大鼠肺泡结构破坏,胶原增生。造模16周数据显示各剂量桑杏护肺颗粒治疗均可有效缓解辐射所致IL-6及TNF-α上升;20周与28周时,高剂量桑杏护肺颗粒组IL-6及TNF-α水平分别下降至阳性对照组水平,但中、低剂量组仍显著高于阳性对照组。结论:高剂量桑杏护肺颗粒可有效减轻大鼠放射性肺损伤,改善继发性纤维沉积,有望作为放射性肺纤维化辅助治疗药物。 Objective:Studying on the therapeutic effect of Sangxing Hufeikeli Granule underwent radiation pulmonary fibrosis in rats.Methods:SD male rats were randomly divided into 6 groups,including controlled group,model group,prednisone group(3.125 mg/kg),Sangxing Hufei Granule group(20 g/kg/d,10 g/kg/d,5 g/kg/d respectively).All rats were given X ray chest single irradiation 14 Gy except the normal group.Each group was executed and acquired lung tissue 32 weeks later.The pathological changes of lung tissue were observed by HE and Masson staining and assessed by Szapiel grade.Meanwhile,the content of serum IL-6 and TNF-α were detected by ELISA assay,which was obtained from all rats’ caudal vein on designated time(4th,8th,12nd,16th,20th,24th,28th,32nd week).Results:Compared with the controlled group,the destroyed lung tissue alveolar structure had been observed in other groups combined with inflammatory cell infiltration and collagen fibers with varying degrees.Meanwhile,the level of IL-6 and TNF-α had been found increased significantly in the experimental groups.Sangxing Hufei Granule exhibited protective effect to the lung of radiation pulmonary pibrosisrats.Fortunately,there was no significant difference between the high dose Sangxing Hufei Granule and Prednisone.Conclusion:Sangxing Hufei Granule would be a potential drug to radiation pulmonary fibrosis.
