摘要
目的探讨益气活血化纤方对肺纤维大鼠肿瘤坏死因子-α(TNF-α)、转化生长因子(TGF-β)、基质金属蛋白酶抑制剂(TIMPs)表达的影响。方法清洁级SD大鼠60只,随机取10只设为空白对照组,剩余50只建立肺纤维化大鼠模型,建模成功后随机分为模型组,强的松组,化纤方高剂量组、化纤方中剂量组和化纤方低剂量组,每组10只。空白对照组和模型组灌服生理盐水,强的松组灌服强的松水溶液,化纤纺织高、中、低剂量组分别按10 g·kg^-1、5 g·kg^-1、2.5 g·kg^-1体质量灌服化纤方水溶液,连续给药28 d,比较各组TNF-α、TGF-β及TIMPs表达水平变化。结果化纤方中剂量组TNF-α、TGF-β及TIMPs水平低于强的松组(P<0.05),强的松组TNF-α、TGF-β及TIMPs水平低于模型对照组(P<0.05);强的松组肺泡部分塌陷,肺泡间隔增厚,炎性细胞浸润轻;化纤方低、高、中剂量组肺泡塌陷不明显,伴有素炎症细胞,肺泡间隔增厚,且化纤方中剂量组症状最轻。结论将中等剂量益气活血化纤方用于肺纤维化大鼠中能降低肺部炎症因子水平,能减轻肺泡炎和肺纤维化程度。
Objective To investigate the effects of Yiqi Huoxue Huaxian Recipe on the expression of tumor necrosis factor-α(TNF-α),transforming growth factor(TGF-β),and matrix metalloproteinase inhibitors(TIMPs)in lung fiber rats.Methods Sixty clean SD rats were randomly selected as blank control groups,and the remaining 50 rats were established as lung fibrosis rat models.After successful modeling,they were randomly divided into model group,prednisone group,and high-dose chemical fiber formula group,chemical fiber prescription middle-dose group and chemical fiber prescription low-dose group,10 rats in each group.The blank control group and model group were perfused with normal saline,the prednisone group was perfused with prednisone aqueous solution,and the chemical fiber textile high,medium,and low dose groups were perfused with the chemical fiber formula at 10 g·kg-1,5 g·kg-1,and 2.5 g·kg-1 respectively.The solution was administered continuously for 28 days,and the expression levels of TNF-α,TGF-β and TIMPs in each group were compared.Results The levels of TNF-α,TGF-βand TIMPs in the middledose group of Huaxian Recipe were lower than those in the prednisone group(P<0.05).The levels of TNF-α,TGF-β and TIMPs in the prednisone group were lower than those in the model control group(P<0.05);the alveolar part of the prednisone group collapsed,the alveolar space thickened,and the inflammatory cells infiltrated lightly;the alveolar collapse of the low-,high-,and medium-dose groups was not obvious,with alveolar inflammation cells,the alveolar space thickened,and the chemical fibers.The middle dose group had the mildest symptoms.Conclusion Applying Yiqi Huoxue Huaxian Reccipe to the lung fibrosis rats can reduce the levels of pulmonary inflammatory factors and reduce the degree of alveolitis and pulmonary fibrosis.
