摘要
目的探讨盐酸法舒地尔联合甲泼尼龙琥珀酸钠干预百草枯中毒致大鼠肺纤维化的效果。方法随机将75只Wistar雄性大鼠分为5组,正常对照组(Cont组)、百草枯中毒组(PQ组)、甲泼尼龙琥珀酸钠干预组(MSS组)、盐酸法舒地尔干预组(FAS组)、甲泼尼龙琥珀酸钠+盐酸法舒地尔干预组(MSS+FAS组),每组各15只。观察并记录各组大鼠日常状况,于第7、14、21天分别处死各组5只大鼠,获取大鼠肺组织标本,用苏木精-伊红染色、Masson染色法观察大鼠第21天肺组织病理学变化,用免疫组化法测定第21天各组大鼠肺组织转化生长因子-β1(TGF-β1)的阳性表达,测定平均光密度值,应用酶联免疫吸附法测定各时间点每组大鼠肺组织匀浆中TGF-β1的含量。结果用FAS、MSS干预PQ中毒大鼠,可减少肺纤维化的程度,FAS联合MSS效果更佳。结论盐酸法舒地尔联合甲泼尼龙琥珀酸钠能够减轻百草枯中毒致大鼠肺纤维的程度。
Objective To investigate the effect of fasudil combined with methylprednisolone sodium succi⁃nate on pulmonary fibrosis induced by paraquat poisoning.Methods Seventy⁃five Wistar male rats were randomly divided into 5 groups,15 rats in each group:normal control group(group Cont),paraquatposoning group(group PQ),methylprednisolone sodium intervention group(group MSS),fasudil intervention group(group FAS),meth⁃ylprednisolone sodium+fasudil intervention group(group MSS+FAS).The daily status of rats in each group was observed and recorded.Five rats in each group were anesthetized and sacrificed on the 7th,14th and 21st day,respectively.The lung tissue samples were obtained.The histopathological changes of rats on the 21st day were observed by hematoxylin⁃eosin and Masson staining.The positive expression of transforming growth factor⁃β1(TGF⁃β1)in the lung tissue of rats in each group on the 21st day was determined by immunohistochemistry.The mean optical density was measured.The content of TGF⁃β1 in the lung homogenates of rats in each group at each time point was determined by enzyme⁃linked immunosorbent assay.Results FAS and MSS could reduce the degree of pulmonary fibrosis,FAS combined with MSS had better effect.Conclusion Fasudil combined with methylprednis⁃olone sodium succinate can reduce the degree of lung fiber induced by paraquat poisoning in rats.
