五味子多糖对NAFLD大鼠肝脏SREBP2/HMGCR表达的影响 被引量:7

摘要 目的探讨五味子多糖(SCP)对非酒精性脂肪肝病(NAFLD)大鼠肝脏胆固醇代谢相关基因变化的影响。方法采用高脂饮食喂养建立NAFLD大鼠模型。40只雄性大鼠,随机分为正常对照组(CON)、正常+SCP组(CON+SCP)、NAFLD模型组(NAFLD)、模型+SCP治疗组(NAFLD+SCP),每组10只,模型组饲喂高脂饲料。4周后,CON+SCP组及NAFLD+SCP组开始灌胃给予SCP 100 mg/kg,CON组及NAFLD组给予等体积溶媒,持续8周。检测血脂水平,RT-PCR和Western Blot方法检测肝脏组织中胆固醇代谢关键因子固醇调节元件结合蛋白(SREBP-2)/3-羟基-3-甲基戊二酸单酰辅酶A还原酶(HMGCR)表达。结果SCP可显著降低NAFLD大鼠血清中TC、TG、LDL-C水平,升高HDL-C水平(P<0.01),并显著降低大鼠肝组织中SREBP-2和HMGCR基因和蛋白的表达。结论SCP可以改善高脂饮食诱导的大鼠NAFLD脂代谢紊乱。其作用机制可能与下调肝脏胆固醇代谢关键因子SREBP-2/HMGCR表达,进而调节肝脏胆固醇代谢平衡有关。 Objective To investigate the effect of Schisandra chinensis polysaccharide(SCP)on liver cholesterol metabolism related genes in rats with nonalcoholic fatty liver disease(NAFLD).Methods NAFLD rat model was established by high-fat diet.Forty male rats were randomly divided into normal control group(CON),normal+SCP group(CON+SCP),NAFLD model group(NAFLD),model+SCP treatment group(NAFLD+SCP),10 rats in each group.After 4 weeks,the CON+SCP group and NAFLD+SCP group were given SCP 100 mg/kg by gavage,and the con group and NAFLD group were given the same volume of solvent for 8 weeks.Blood lipid levels were measured.The expression of SREBP-2/HMGCR in liver tissue was detected by RT-PCR and Western blot.Results SCP significantly decreased the levels of TC,TG and LDL-C in serum,increased the level of HDL-C(P<0.01),and significantly reduced the expression of SREBP-2 and HMGCR genes and proteins in liver tissue of NAFLD rats.Conclusion SCP can improve the lipid metabolism disorder of NAFLD induced by high-fat diet.The mechanism may be related to down regulating the expression of SREBP 2/HMGCR,the key factor of liver cholesterol metabolism,and then regulating the balance of liver cholesterol metabolism.
