黄芩苷对乳腺癌MCF-7/dox细胞注射用盐酸多柔比星化疗敏感性的影响 被引量:2

Effect of baicalin on adriamycin chemosensitivity of breast cancer MCF-7/dox cells

摘要 目的研究黄芩苷对乳腺癌MCF-7/dox细胞注射用盐酸多柔比星化疗敏感性的影响及机制。方法取对数生长期的MCF-7、MCF-7/dox细胞,预实验分析黄芩苷最佳干预浓度,分为10,20,30,40,50,80,100,150,200 mg·L^-1黄芩苷干预组和对照组(等量生理盐水干预)。设置实验组(3.68μmol·L^-1注射用盐酸多柔比星+40 mg·L^-1黄芩苷)和对照组(3.68μmol·L^-1注射用盐酸多柔比星),处理24 h。以噻唑蓝(MTT)法检测细胞增殖情况,以流式细胞术检测MCF-7、MCF-7/dox细胞凋亡情况;以蛋白质印迹法检测MCF-7、MCF-7/dox细胞Beclin-1、微管轻链Ⅰ蛋白3-Ⅱ型(LC3Ⅱ)的表达。结果黄芩苷50 mg·L^-1及以上浓度干预后MCF-7、MCF-7/dox细胞增殖率均低于对照组,并且随着浓度增大,细胞增殖率逐渐降低(P<0.05)。用40 mg·L^-1黄芩苷对注射用盐酸多柔比星干预组进行后续实验。对照组和实验组MCF-7细胞增殖率分别为(45.68±5.32)%,(41.26±2.05)%;MCF-7/dox细胞增殖率分别为(73.63±2.48)%,(68.72±2.81)%。与对照组相比,细胞增殖率均降低(P<0.05);与对照组相比,黄芩苷将MCF-7细胞及MCF-7/dox细胞对注射用盐酸多柔比星的化疗敏感性显著提高(P<0.05);与对照组比较,实验组MCF-7、MCF-7/dox细胞早期凋亡比例及Beclin-1、LC3Ⅱ蛋白相对表达量均显著升高(P<0.05)。结论黄芩苷可通过上调Beclin-1、LC3Ⅱ蛋白表达增强细胞自噬水平发挥对乳腺癌注射用盐酸多柔比星化疗的增敏作用。 Objective To investigate the effect and mechanism of baicalin on the sensitivity of breast cancer MCF-7 dox cells to adriamycin chemotherapy.Methods MCF-7 and MCF-7/dox cells in logarithmic growth phase were taken.Pre-experiment analyzed of the optimal concentration of baicalin intervention,baicalin intervention group(10,20,30,40,50,80,100,150,200 mg·L^-1 baicalin intervention)and control group(equivalent saline intervention).Test group(3.68μmol·L^-1 doxorubicin hydrochloride for injection+40 mg·L^-1 baicalin)and control group(3.68μmol·L^-1doxorubicin hydrochloride for injection)were set up,Methyl thiazolyl tetrazolium(MTT)method was used to detect cell proliferation after 24 h of intervention.Annexin V-FITC/PI double staining method was used to detect MCF-7 and MCF-7/dox cell apoptosis.Western blot method was used to detect the expression of Beclin-1,and microtubule light chain protein type 3-Ⅱ(LC3Ⅱ)in the MCF-7,MCF-7/dox cells.Results The proliferation rate of MCF-7 and MCF-7/dox cells after intervention of baicalin50 mg·L^-1 and above was lower than that of control group and with the increase of baicalin concentration,MCF-7 and MCF-7/dox The cell proliferation rate gradually decreased(P<0.05).In this study,40 mg·L^-1 baicalin which was interfered by doxorubicin hydrochloride for injection was used for follow-up experiments.The proliferation rates of MCF-7 cells in control and test groups were(45.68±5.32)%,(41.26±2.05)%;the MCF-7/dox cell proliferation rates were(73.63±2.48)%,(68.72±2.81)%.Compared with control group,the proliferation rate of MCF-7 and MCF-7/dox cells in test group were all reduced(all P<0.05);baicalin increased the sensitivity of MCF-7 cells and MCF-7/dox cells to adriamycin(P<0.05).Compared with control group,the early apoptosis ratio of MCF-7,MCF-7/dox cells and the relative expression of Beclin-1 and LC3Ⅱprotein in test group were all significantly increased(all P<0.05).Conclusion Baicalin can enhance the cell autophagy level by increasing the expression of Beclin-1 and LC
