乳结消颗粒对家兔乳腺增生的影响 被引量:2

Effects of Rujiexiao Granules on rabbit with mammary glands hyperplasia

摘要 目的:观察乳结消颗粒对乳腺增生病家兔模型的药理作用。方法:通过给健康雌性未孕家兔臀部交替肌肉注射苯甲酸雌二醇和黄体酮,建立乳腺增生模型。模型建立成功后,将家兔随机分为7组(每组8只):正常对照组,模型组,乳结消高、中、低剂量(6、3、1.5g生药/kg)组,乳疾灵组(4g/kg),三苯氧胺组(1.8mg/kg),各组灌胃给药,每日1次,连续给药30d,正常对照组和模型组家兔每日灌胃等体积蒸馏水。于造模前、造模30d、给药15d及给药30d,采用卡尺测量乳头直径和高度,天平称量乳腺、子宫、脾脏及胸腺重量,容积法测量乳腺体积,ELISA法测定性激素水平,化学法测定超氧化物歧化酶(SOD)和丙二醛(MDA),乳腺组织进行病理形态观察等。结果:乳结消颗粒明显减少模型家兔乳头直径、高度及乳腺体积,减轻乳腺重量,减少乳腺小叶数和小叶内腺泡数,减小腺泡腔直径和导管腔直径;降低血清雌二醇(E2)水平,增加血清孕酮(P)含量;增加SOD活性;抑制子宫肿大和胸腺萎缩。结论:乳结消颗粒具有调节性激素水平,增强清除氧自由基的能力,减轻乳房肿大,减轻乳腺组织增生性病变等作用,这为临床应用该药治疗乳腺增生病提供了实验依据。 Objective:To observe the pharmacological effects of Rujiexiao Granules on rabbit with mammary glands hyperplasia.Methods:To establish the model of mammary glands hyperplasia by intramuscular injection of estradiol benzoate and progestin to non-pregnant female rabbit.After modeling,rabbits were randomly divided into 7 groups(8 per group):control group,model group,high-dose,middle-dose and low-dose of Rujiexiao Granules group(6 g,3 g,and 1.5 g crude drug/kg,respectively),Rujiling Granules group(4 g/kg),and tamoxifen group(1.8 mg/kg).Intragastric administration was conducted once a day for another 30 days,while rabbits in control group and model group were given equivalent distilled water.Diameter and hights of nipples were measured by caliper,weights of mammary,uteri and thymus glands were measured by balance,volumes of mammary were measured through volume method,hormone levels were measured through ELISA,SOD and MDA were measured through chemical method,pathomorphism observations of mammary were conducted before modeling,30 days after modeling,15 days and 30 days after treatment.Results:The papilla diameter,mammary gland volume and mammary gland weight were significantly reduced in the Rujiexiao Granules groups.The number of mammary gland lobules,number of acinar glands in lobules,diameter of acinargland cavities and diameter of acinar gland ducts were reduced in the Rujiexiao Granules groups.The serum estradiol(E2)were reduced,while the serum progesterone(P)level and SOD activity was increased significantly and the uterine enlargement and thymus atrophy were suppressed in the Rujiexiao Granules groups.Conclusion:Rujiexiao Granules has effects of regulating hormone level,enhancing the ability of oxygen free radicals elimination,reducing breast swelling and hyperplasia in mammary glands,which provides experimental evidence for clinical application.
