摘要
目的基于“乳为肝窍”理论,探讨柴胡疏肝散对高泌乳素血症大鼠乳腺组织及性激素的作用,探寻肝乳相关的现代科学依据。方法将48只大鼠随机分为正常组8只和造模组40只,造模组大鼠采用皮下注射灭吐灵方法制成高泌乳素血症模型。将造模成功大鼠随机分为模型组、溴隐亭组、柴胡疏肝散高剂量组、柴胡疏肝散中剂量组、柴胡疏肝散高剂量组,每组8只。溴隐亭组灌胃溴隐亭水溶液2 mg/(kg·d),柴胡疏肝散低、中、高剂量组分别灌胃柴胡疏肝散药液3.15 g/(kg·d)、6.3 g/(kg·d)、12.6 g/(kg·d),正常组和模型组灌胃等容量生理盐水,均每日1次,连续30 d。末次灌胃后,腹腔注射麻醉大鼠,测量各组大鼠第3对乳头直径、高度,取血检测血清雌二醇(E2)、卵泡生成激素(FSH)、泌乳素(PRL)水平,取第3对乳腺组织进行病理观察。结果模型组大鼠乳头直径、乳头高度及血清PRL水平均明显高于正常组(P均<0.05),血清E2和FSH水平均明显低于正常组(P均<0.05);溴隐亭组及柴胡疏肝散中、高剂量组大鼠乳头直径、乳头高度及血清PRL水平均明显低于模型组(P均<0.05),血清E2和FSH水平均明显高于模型组(P均<0.05)。模型组大鼠乳腺组织呈弥漫性增生,乳腺导管明显扩张,部分导管上皮细胞呈分泌前期改变;柴胡疏肝散高剂量组大鼠乳腺组织增生情况较模型组明显缓解。结论柴胡疏肝散可改善乳腺组织增生性、泌乳性病变,纠正高泌乳素血症大鼠血清性激素紊乱。血清性激素受肝脏代谢功能的影响,作用于乳腺所产生的生理、病理变化,可能是肝乳相关学说的现代调控依据之一。
Objective It is to discuss the effect of Chaihu Shugan Powder(CSP)on the mammary tissues and the sex hormones of rats with hyperprolactinemia(HPRL)on the theory of“the Nipple,the Liver’s Organ”,and to seek the contemporary scientific evidences that relate to liver and breast.Methods Forty-eight rats were randomly divided into normal group(n=8)and molding group(n=40),the rats in the molding group were given paspertin by subcutaneous injection to establish HPRL models.The successful models of rats were randomly divided into model group,bromocriptine group,high-dose CSP group,medium-dose CSP group,and high-dose CSP group,8 rats in each group.The bromocriptine group was gavaged with bromocriptine solution 2 mg/(kg·d),and the low,medium and high dose CSP groups were gavaged with CSP 3.15 g/(kg·d),6.3 g/(kg·d),12.6 g/(kg·d),respectively,the normal group and the model group were given the same volume of normal saline,once a day for 30 consecutive days.After the last intragastric injection,the rats were anesthetized by intraperitoneal injection,the diameter and height of the third pair of nipples in the rats of each group were measured,and their blood was taken to detect the levels of serum estradiol(E2),follicle-producing hormone(FSH),and prolactin(PRL),the pathological changes of the third pairs of breast tissue was observed.Results The nipple diameter,nipple height,and the level of serum PRL in the model group were significantly higher while the levels of serum E2 and FSH were significantly lower than those in the normal group(all P<0.05);the nipple diameter,nipple height,and the level of serum PRL in the bromocriptine group and medium and high dose CSP groups were significantly lower while the levels of serum E2 and FSH were significantly higher than those in the model group(all P<0.05).The mammary gland tissue of the model group showed diffuse hyperplasia,the mammary gland ducts were significantly expanded,and some ductal epithelial cells showed pre-secretory changes.The hyperplasia of the mammary gland ti
