HPLC-DAD法测定狭叶金粟兰不同部位中绿原酸、伞形花内酯和异嗪皮啶的含量 被引量:1

摘要 目的建立HPLC-DAD法测定狭叶金粟兰不同部位(根、茎、叶)中绿原酸、伞形花内酯和异嗪皮啶的含量,为金粟兰科药用植物狭叶金粟兰的质量控制和资源深入开发提供参考依据。方法色谱柱为Phenomenex C 18柱(250 mm×4.6 mm,5μm);流动相为乙腈(A)-0.1%磷酸溶液(B),按梯度洗脱程序进行洗脱:0~9 min,12%~20%A;9~15 min,20%~25%A;15~25 min,25%~35%A;25~45 min,35%~45%A;45~60 min,45%~20 A;流速:1.0 ml/min;检测波长330 nm;柱温30℃;进样量20μl。结果绿原酸、伞形花内酯和异嗪皮啶分别在6.1447~36.868μg/ml(r=0.9994)、0.2787~1.6720μg/ml(r=0.9991)和0.9047~5.4280μg/ml(r=0.9992)范围内与峰面积呈良好的线性关系,精密度、重复性及稳定性均良好,绿原酸、伞形花内酯和异嗪皮啶平均回收率分别为99.06%(RSD=1.49%)、99.32%(RSD=1.35%)、99.67%(RSD=1.61%)。狭叶金粟兰不同部位中绿原酸、伞形花内酯和异嗪皮啶的含量次序为:根>茎>叶。结论本试验建立的方法简便快捷、准确可靠,重现性高,可为狭叶金粟兰的质量控制提供依据。狭叶金粟兰不同部位中均含有绿原酸、伞形花内酯和异嗪皮啶,但其含量差异较大,其根中绿原酸、伞形花内酯和异嗪皮啶含量最高,在临床使用和资源开发时可以予以区别。 Objective To establish the HPLC-DAD method to determine the content of chlorogenic acid,umbelliferone and isofraxidin from different parts of Chloranthus angustifolius Oliv,and provide references for the quality control of chloranthus angustifolius Olive and further development and utilization of its resources.Methods HPLC-DAD was adopted with Phenomenex C 18(250 mm×4.6 mm,5μm)column.The mobile phase was acetonitrile(A)-0.1%phosphoric acid(B)solution with gradient elution:0~9 min,12%~20%A;9~15 min,20%~25%A;15~25 min,25%~35%A;25~45 min,35%~45%A;45~60 min,45%~20 A.The flow rate was 1.0 ml/min,detection wavelength was 330 nm,column temperature was 30℃,and the injection volume was 20μl.Results The concentration of chlorogenic acid,umbelliferone and isofraxidin had good linear relationship with peak area within the range of 6.1447~36.868μg/ml(r=0.9994),0.2787~1.6720μg/ml(r=0.9991),and 0.9047~5.4280μg/ml(r=0.9992).The precision,repeatability and stability were all good,and the average sample recovery was 99.06%(RSD=1.49%),99.32%(RSD=1.35%),and 99.67%(RSD=1.61%),respectively.The content of chlorogenic acid,umbelliferone and isofraxidin from different parts of Chloranthus angustifolius Oliv were as follows:root﹥stem﹥leaf.Conclusion The established method is simple,rapid,accurate and reliable with high reproducibility,which can provide basis for the quality control of Chloranthus angustifolia Oliv.Chlorogenic acid,umbelliferone and isofraxid is found in different parts of Chloranthus angustifolia Oliv,but the content of chlorogenic acid,umbelliferone and isefraxid in its root is the highest,which can be distinguished in clinical use and resource development.
