白芥子及炒白芥子饮片、水煎液、配方颗粒UPLC特征图谱研究 被引量:4

摘要 目的:采用UPLC法对白芥子及炒白芥子饮片、水煎液和配方颗粒的特征图谱进行研究,考察各工艺过程中主要特征峰的变化情况。方法:采用WATERS CORTECS T3色谱柱(150 mm×2.1 mm,1.6μm),流动相为甲醇-0.1%磷酸,梯度洗脱,流速为0.25 mL/min,柱温为30℃,检测波长为278 nm,利用国家药典委员会颁布的"中药色谱指纹图谱相似度评价系统2012A版"对色谱图进行匹配。结果:分别建立了白芥子及炒白芥子饮片、水煎液、配方颗粒的UPLC特征图谱。白芥子饮片特征图谱确定了13个共有峰,其中11个共有峰可在水煎液和配方颗粒中得到追踪;炒白芥子饮片特征图谱确定了11个共有峰,11个共有峰均可在水煎液和配方颗粒中得到追踪。结论:建立的UPLC特征图谱能够快速有效区分白芥子与炒白芥子饮片、水煎液、配方颗粒,可为白芥子与炒白芥子炮制品质量控制、配方颗粒提取生产过程质量控制及成品质量标准制定提供依据。 Objective: To study the characteristic spectra of decoction pieces, water decoctions and formula granules between Baijiezi(sinapis albae semen) and Chaobaijiezi(sinapis albae semen frictum) by UPLC method,and to investigate the changes of the main characteristic peaks in each process. Methods: The analysis was carried out by a WATERS CORTECS T3 column(150 mm×2.1 mm,1.6 μm) with methanol-0.1% phosphoric acid as the mobile phase in a gradient elution. The detection wavelength was 278 nm with a flow rate of 0.25 mL/min. The column temperature was 30 ℃. The chromatograms were matched by the similarity evaluation system of chromatographic fingerprint of traditional Chinese medicine(2012 a) issued by the National Pharmacopoeia Committee. Results: The UPLC characteristic spectra of decoction pieces, water decoctions and formula granules, from 10 batches of Baijiezi(sinapis albae semen) and Chaobaijiezi(sinapis albae semen frictum), were established respectively. The common peaks of UPLC characteristic spectra from 10 batches of Baijiezi(sinapis albae semen) and Chaobaijiezi(sinapis albae semen frictum) decoction pieces were 13 and 11, respectively, and 11 common peaks in both of the UPLC characteristic spectra could be tracked in the water decoctions and formula granules. Conclusion: The UPLC characteristic spectra can distinguish decoction pieces, water decoctions and formula granules between Baijiezi(sinapis albae semen) and Chaobaijiezi(sinapis albae semen frictum) quickly and effectively, and provide a basis for quality control of processing of Baijiezi(sinapis albae semen), extraction and production of formula granules and formulation of products quality standards.
