复方金银花颗粒UPLC指纹图谱研究及3种成分含量测定 被引量:2

摘要 目的:建立复方金银花颗粒UPLC指纹图谱,并测定其中3种主要成分含量,为复方金银花颗粒质量控制提供部分参考。方法:采用AcquityUPLCBEHC18柱(2.1mm×100mm,1.7μm)色谱柱,流动相为甲醇-0.1%磷酸水,梯度洗脱,流速为0.30mL/min,柱温为30℃,检测波长为320nm。结果:建立了复方金银花颗粒专属性的UPLC指纹图谱,确定了16个共有峰并测定了其中绿原酸、黄芩苷、连翘苷3种成分含量,15批复方金银花颗粒制剂相似度均大于0.97;聚类分析可将15批产品分为2类;绿原酸、栀子苷和黄芩苷分别在18~180、10~100、21~210μg/mL浓度范围内线性关系良好,平均回收率在97.95%~99.26%。结论:该方法稳定可靠,可用于复方金银花颗粒的质量控制与评价。 Objective:To establish the UPLC fingerprint of Compound Lonicera Granules and determine the contents of three main components,and provides some reference for the quality control of Compound Lonicera Granules.Methods:The separation was performed on Acquity UPLC BEH C18(2.1 mm×100 mm,1.7μm)column gradient eluted with the mobile phase consisting methanol and 0.1%phosphoric acid water in gradient mode.The flow rate was 0.30 mL/min and the column temperature was 30℃.The detection wavelength was set at 320 nm.Results:The UPLC fingerprint of 15 batches of Compound Lonicera Granules were established and it had16 characteristic common peaks.The contents of three components of chlorogenic acid,baicalin and forsythin were determined.The similarity degrees of 15 batches of the sample were more than 0.97.The 15 batches of sample were classified into two categories through pattern recognition.Chlorogenic acid,baicalin and phillyrin showed a good linearity in 18-180μg/mL,10-100μg/mL and 21-210μg/mL,respectively.The average recoveries were 97.95%-99.26%.Conclusion:The method is stable and reliable,and can be used for the quality control and evaluation of Compound Lonicera Granules.
