摘要
蔗茅(Erianthus fulvus Ness.)是甘蔗亚族(Subtrib.Saccharinae)、蔗茅属(Erianthus Michaux.)内的一个野生种,具有较强的耐寒特性。为挖掘和利用蔗茅耐寒基因,本研究在转录组测序的基础上利用电子克隆结合RT-PCR的技术从"蔗茅99-3"无性系中克隆到一个WRKY家族基因,该基因的开放阅读框拥有358个编码氨基酸,CDS全长1077 bp,分子量为37.4 kD,MH709119);荧光定量PCR检测结果显示,低温胁迫下,该基因在2个蔗茅(蔗茅无性系I91-8和蔗茅无性系99-3)和2个甘蔗栽培品种/系(ROC10和滇蔗04-14)的苗期均表现出上调状态。研究结果可为耐寒、适应性广的甘蔗新品种(系)的培育提供理论依据和技术支撑。
Erianthus fulvus Ness.is a wild species closely related to sugarcane and has strong cold resistance.In this study,based on transcriptome sequencing,a WRKY family gene was cloned from"sugarcane arcane 99-3"clone using electronic cloning combined with RT-PCR.The open reading frame of this gene has 358 coding amino acids,the full length of CDS is 1077 bp,the molecular weight is 37.4 kD,the isoelectric point is 6.24,and the average pro-level coefficient is-0.380.By homologous sequence alignment,the gene was named EfWRKY62(NCBI login number:MH709119).Fluorescence quantitative PCR results showed that the gene was up-regulated at the seedling stage of two clones of E.fulvus(sugarcane I91-8 and sugarcane 99-3)and two sugarcane cultivars/lines(ROC 10 and Dianzhe 04-14)under low temperature stress.To provide a theoretical basis and technical support for excavating and utilizing the cold-tolerant genes of the E.fulvus Ness.,and then breeding a new variety of sugarcane varieties(lines)with wide adaptability.
