重组人骨形成蛋白-4对人成骨样细胞MG-63增殖分化的影响

摘要 目的:探讨重组人骨形成蛋白-4(rhBMP-4)对人成骨样细胞MG-63的增殖分化的影响。方法:人成骨样细胞系MG-63用含10%胎牛血清的DMEM高糖培养基培养在5%CO2、37℃培养箱中,rhBMP-4诱导后用MTT比色法测定细胞增殖,用PNP比色法测定细胞碱性磷酸酶活性,羟脯氨酸测定试剂盒测定细胞羟脯氨酸含量。结果:5、10、20 ng/mL的rhBMP-4均能诱导MG-63细胞的增殖(P<0.05),且呈浓度及时间依赖性;10 ng/mL rhBMP-4能增强MG-63细胞的碱性磷酸酶活性及Ⅰ型胶原的分泌(P<0.05)。结论:rhBMP-4能诱导人成骨样细胞MG-63的增殖及分化。 Objective:To investigate the effect of recombinant human bone morphogenetic protein-4(rhBMP-4 on the proliferation and differentiation of human osteoblast-like cells MG-63.Methods:Human osteoblast-like cell line MG-63 was cultured in DMEM high-glycemic medium containing 10%fetal bovine serum in a 5%CO2,37°C incubator,induced with rhBMP-4,and cell proliferation was measured by MTT colorimetry.The PNP colorimetric method was used to determine the cell alkaline phosphatase activity,and the hydroxyproline determination kit was used to determine the cell hydroxyproline content.Results:5,10,and 20 ng/mL rhBMP-4 could induce the proliferation of MG-63 cells in a concentration and time-dependent manner(P<0.05).10 ng/mL rhBMP-4 could enhance the alkaline phosphatase activity and typeⅠcollagen secretion of MG-63 cells(P<0.05).Conclusion:rhBMP-4 can induce the proliferation and differentiation of human osteoblast like cells MG-63.
