水苏碱对异丙肾上腺素致小鼠心肌纤维化的保护作用 被引量:2

摘要 目的观察益母草提取物水苏碱对小鼠心肌纤维化(myocardialfibrosis,MF)的保护作用。方法小鼠50只,随机分为5组(n=10):对照组、模型组、水苏碱高剂量组、水苏碱中剂量组、水苏碱低剂量组。模型组,水苏碱高、中、低剂量组每只皮下注射盐酸异丙肾上腺素2.5 mg/kg造模,持续28 d,对照组按照2.5 mg/kg皮下注射等量生理盐水。水苏碱高、中、低剂量组造模同时分别腹腔注射10.0、5.0、2.5 mg/kg水苏碱,对照组、模型组腹腔注射等量生理盐水,连续28 d。测定小鼠心重指数、血清中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、心肌组织羟脯氨酸含量,HE和Masson染色分析心肌组织病理改变。结果与对照组相比,模型组心重指数明显增加,SOD活性显著性下降,MDA含量显著升高,羟脯氨酸含量明显增加;与模型组比较,水苏碱高、中、低剂量组小鼠心重指数明显下降,SOD活性显著提高,MDA含量明显下降,羟脯氨酸含量明显降低,心肌组织病理学改变缓解。结论水苏碱能有效改善异丙肾上腺素诱发的MF,其机制可能与提高SOD活性、降低MDA含量有关。 Objective:To observe the protective effect of stachydrine on myocardial fibrosis(MF)in mice.Methods:Fifty mice were randomly divided into five groups(n=10):control group,model group,stachydrine high dose group,medium dose group and low dose group.Mice in the model group and the administration group were subcutaneously injected with isoprenaline hydrochloride(Iso)2.5mg/kg for 28 days.Stachydrine(10.0,5.0,2.5 mg/kg)was injected intraperitoneally in the high,middle and low dose group for 28 days.The heart weight index,the activity of superoxide dismutase(SOD),the content of MDA and the content of hydroxyproline in myocardium were measured.The pathological changes of myocardium were analyzed by HE and Masson staining.Results:Compared with the control group,the heart weight index of the model group increased,SOD activity decreased significantly,MDA content increased,and hydroxyproline content increased.Compared with the model group,the heart weight index decreased significantly,SOD activity increased,MDA content decreased,hydroxyproline content decreased,and myocardial histopathological changes relieved in the stachydrine treated group.Conclusion:Stachydrine can effectively improve the MF induced by Iso,and its mechanism may be related to the increase of SOD activity and the decrease of MDA content.
