苦参碱治疗包虫病后小鼠免疫功能和部分氧化还原酶、转氨酶的变化及意义 被引量:10

摘要 目的考察用药后小鼠的免疫功能变化并研究对小鼠肝组织损伤和棘球蚴组织营养代谢产生影响的部分氧化还原酶、转氨酶等的改变状况。方法染病小鼠经苦参碱、阿苯达唑及联合用药治疗90天后,对血清中Th2型细胞因子IL-4,IL-6和IL-10含量,及T淋巴细胞亚群分类进行检测;采用比色法、放射免疫标记技术及速率法等检测治疗前后小鼠肝脏、棘球蚴囊壁组织匀浆NO、NOS、LD、LDH和AKP及血清HA、GOT的含量及活性。结果治疗后血清中IL-4,IL-6和IL-10的含量降低(P<0.05),其中IL-4与IL-10在联合用药组下降尤为明显(P<0.05);小鼠外周血中T细胞亚群分类:CD4+/CD8+升高(P<0.05),但与正常小鼠比较尚存在差异(P<0.05)。棘球蚴囊壁LD、LDH和AKP均升高(P<0.05),联合用药组尤为明显(P<0.05)。肝脏组织中各用药组NO、NOS和下降(P<0.05),LDH在苦参碱组及联合用药组下降幅度明显大于Alb组(P<0.05);LD仅在苦参碱组与联合用药组下降(P<0.05)。各用药组血清HA含量较之对照组明显降低(P<0.05),且苦参碱组及联合用药组与Alb组有显著性差异(P<0.05)。苦参碱组及联合用药组血清GOT含量较之对照组降低(P<0.05),Alb组无变化(P>0.05)。结论棘球蚴病小鼠经治疗后,机体的免疫状况有所恢复。苦参碱和阿苯达唑均能抑制棘球蚴影响代谢,尤其联合应用效果更好。苦参碱能显著改善小鼠肝功能,并对逆转肝细胞损伤方面较阿苯达唑为优。 In this study the alterations of immune functions and the changes of certain redox enzymes and transaminase in liver tissue and echinococcus cysts of mice infected with echioncoccus granulosus after treatment with matrine(Mat) and albendazole(Alb) were investigated. The echinococcus mouse models established were treated with Mat, Alb alone or in combination with this two drugs for 90 days, and the levels of IL-4, IL-6 and IL-10 in serum were detected through radioimmunoassay (RIA) before and after treatment; as well as the CD^+4 and CD^+8 T cells from PMBC classified by flow cytometry (FCM), content and activity of HA and GOT in serum measured by RIA and velocity respectively, the NO and LD content and NOS, LDH activity in hepatic tissue homogenate from echinococcus infected mice and the LD content, LDH and AKP activity in Echinococcus granulosus cyst wall measured by chromatometry. The experimental results demonstrated that the Th2 type cytokines IL-4 , IL-6 and IL-10 were decreased after treatment, especially the decrease of IL-4 and IL-10 in the case of the combined treatment with Mat and Alb. And the alterations in T lymphocyte subsets were obvious with an increase of CD^+8 T cells and a concomitant decrease of CD^+8 T cells, resulting an elevation of the CD^+4/CD^+8 ratio. However, the differences between normal and the treated mice still existed. In the echinococcus cysts walls,the content of LD and the activitiy of LDH and AKP were all increased, especially more obvious in the case of the combined treatment with both drugs, in which the magnitude of decreased. LDH in Mat-treated group was more prominent than that in the Alb-treated group, and LD just decreased only in the Mat-treated group and the combined treatment group. While in the hepatic tissue homogenates, the NO content and the NOS activity decreased in all groups of the treated mice. The serum HA content was decreased apparently in the experimental groups in comparison with the control group, but significant differences
