摘要
本文采用棘球蚴囊壁生发层制备抗原免疫BALB/c小鼠脾细胞与骨髓瘤SP2/O进行细胞融合,经多次筛选克隆后建立了分泌抗细粒棘球蚴单克隆抗体细胞株C_5F_2C_(11)A_(11),C_(12)C_(11)F_6G_3,C_(12)H_(12)F_8D_5三株。应用ELISA及免疫组化法检测该三株具有特异性,培养上清分泌抗体滴度分别为1:2560,1:5120,1:5120。注入同系小鼠腹腔诱生肿瘤产生单抗滴度分别为1:1024000,1:4096000,1:2048000。杂交瘤细胞株经组织培养传代,分泌的抗体性能稳定。分泌抗细粒棘球蚴抗体杂交瘤细胞株的建立,对包虫病的免疫学诊断和治疗具有重要的价值和意义。
The spleen cells from the BALB/c mice immunized by using germinal layer antigen ofEchinococcus granulosus were fused with the myeloma SP2/O cell line. We got three hybridoma cell lines pro-ducing specific antibody named C5F5C11 A11、C12C11 F6G3 and C12H12F8D5,respectively by repeatedly screeningand cloning after fusion. Using ELISA and immunohistochemical method,these cell lines have been identifiedthat they can secrete the monoclonal antibody against the germinal layer antigen of Echinococcus granulosus. The antibody titers in the supernates of the cell cultures were 1:2 560、1:5 120 and 1:5 120 respec-tively.The cell lines were infected into the abdominal cavity of congeneric BALB/c mice to induce the hy-bridoma lines which can secret the McAb against the germinal layer antigen. The titers of the McAb producedby the hybridoma cells were 1:1 024 000、1:4 096 000 and 1:2 048 000 respectively. The hybridoma celllines were reproduced by tissue culture.The secreted McAb had high and stable activity. The establishment of the McAb cell lines has important value and significance for immunological diagno-sis and treatment of hvdatid disease.
