摘要
目的:考察白茎蓝刺头和新疆蓝刺头不同部位芦丁和总黄酮的含量。方法:采用水提取法、乙醇提取法对白茎蓝刺头化学成分进行定性试验;HPLC法对其中的芦丁含量进行测定:色谱柱为Agilent C18(250mm×4.6mm,5mm),流动相为甲醇-0.05%磷酸溶液(37∶63),流速为1.0m L·min-1,检测波长360nm,柱温30℃;采用紫外分光光度法,以芦丁为对照品,三氯化铝显色后在波长508nm处测定白茎蓝刺头及新疆蓝刺头不同部位总黄酮含量。结果:白茎蓝刺头含有黄酮类、生物碱、三萜类等特征化学成分;芦丁的线性范围为13.06~78.37mg·m L^-1,R2=0.9998,平均回收率为98.26%(RSD=1.74%,n=6);总黄酮的线性范围为0~48.71mg·m L^-1,R2=0.9999,平均回收率为100.28%(RSD=1.73%,n=6),结论:白茎蓝刺头中总黄酮含量为花>叶>全草>根>茎;新疆蓝刺头中含量为花>叶>全草>根>茎。
Objective:To study the contents of total flavonoids and Rutin in different parts of Echinops albicaulis and Echinops ritro L.were determined.Methods:The methods of water extraction and ethanol extraction were used.The detection was carried out by HPLC using Agilent C18,column(250mm×4.6mm,5mm)was used with methanol-0.05%phosphoric acid solution(37∶63)as the mobile phase;the detection wavelength was360nm,the flow rate 1.0 ml·min-1and the column temperature was 30℃.UV spectrophotometry was adopted to determine the content of total flavonoids,using the Al Cl3,coloration method with rutin reference to measure at 508 nm wave 1 engths.Results:Echinops albicaulis possibly contains chemical constituents including alkaloid,flavones,triterpene compounds etc.the results showed the standard curve of falvonoids was linear in the range of13.06~78.37mg·m L^-1,(R2=0.9998),The average recovery was 98.26%(RSD=1.74%,n=6),the standard curve of the rutin was linear in the range of 0~48.71mg·m L^-1,(R2=0.9999).The average recovery was 100.28%(RSD=1.73%,n=6).Conclusion:The content of total flavonoids in the different parts of Echinops albicauris sequence is flower>leaf>whole grass>root>stem.The contentin Echinops ritro L.was flower>leaf>whole grass>root>stem.
