摘要
目的建立蓝刺头花中总黄酮的含量测定方法,研究蓝刺头花中总黄酮的提取工艺。方法采用乙醇回流提取法,以亚硝酸钠-硝酸铝-氢氧化钠为试剂、芦丁为对照测定蓝刺头花中总黄酮的含量。在单因素试验的基础上,采用L9(3~4)正交试验设计,研究乙醇浓度、料液比、提取时间和提取次数对蓝刺头花总黄酮得率的影响。结果蓝刺头花中总黄酮最佳提取工艺条件为:乙醇浓度95%、提取次数3次、每次回流时间0.5 h、料液比为1∶30(g/m L),在最佳条件下的黄酮含量为50.29 mg·g^(-1)。结论优选得到的工艺简单、稳定、可行、重复性好,为蓝刺头花的进一步开发利用提供依据。
Objective To establish a reliable method to determine total flavoniods in Echinops transiliensis flowers, and to determine the extraction conditions. Methods Flavonoids was extracted from Echinops transiliensisflowers, with ethanol as the solvent, sodiunl nitrite, aluminium nitrate and sodium hydroxide as the indicators, and rutin as the control sample. Based on single factor test, the effects of ethanol concentration, extraction time, solid/liquid ratio(g/m L) and extraction times on the extraction rate of total flavonoids were studied according to L9(34) orthogonal test. Results The optimum extraction conditions were: 95% ethanol, 1∶30 solid-liquid ratio, 30 min extraction and extraction for 3 times. Under the above extraction conditions, the content of total flavonoid reached 50.29 mg·g^-1. Conclusion This optimized process for total flavonoids from Echinops transiliensis flower is simple, stable, feasible and repeatable, and provids references for its further research and application.
