摘要
目的:建立小毛茛内酯的含量测定方法,优选猫爪草中小毛茛内酯的提取工艺。方法:采用HPLC测定小毛茛内酯含量,流动相甲醇-乙腈-水(40∶40∶20),检测波长254 nm。在单因素试验基础上,以小毛茛内酯质量分数为指标,采用L9(34)正交试验考察提取时间、料液比、乙醇体积分数及粉碎粒度对提取工艺的影响。结果:最佳提取工艺为粉碎粒度80目,加3倍量70%乙醇提取3次,每次2 h;小毛茛内酯质量分数(1.24±0.22)%。结论:优选的工艺稳定可行且提取率高,为猫瓜草的开发与利用提供参考。
Objective: To establish a method for determination of ternatolide and optimize its ultrasonic extraction process from Ranunculi Ternati Radix. Method: HPLC was adopted to determine the content of ternatolide with mobile phase of methanol-acetonitrile-water (40: 40: 20) and detection wavelength at 254 nm. Based on single-factor tests, orthogonal design was used to optimize extraction technology by taking extraction time, ratio of solid to liquid, the concentration of ethanol and grinding particle size as factors. Result: Optimal ultrasonic extraction conditions were as follows: extracted thrice with three times the amount of 70% ethanol for 2 h each time, grinding particle size of 80 mesh; under these conditions, the content of ternatolide was ( 1.24 ± 0.22) %. Conclusion: Optimized extraction process was stable and feasible with high extraction yield, this study could provide a reference for development and utilization of Ranunculi Ternati Radix.
