线粒体氧化磷酸化对肺腺癌细胞A549迁移影响的研究 被引量:1

摘要 通过MTT法确定抗霉素A(线粒体功能抑制剂)、三溴丙酮酸(糖酵解功能抑制剂)及丙酮酸钠的最佳药物浓度,对体外培养的A549肺腺癌细胞进行处理,利用组织化学法和划痕实验法检测了线粒体氧化磷酸化在肺腺癌细胞迁移过程中的作用.结果表明:利用MTT法确定的抗霉素A、三溴丙酮酸及丙酮酸钠的最佳药物浓度分别为150μmol/L,200μmol/L和0.02 mol/L;利用组织化学法及划痕实验法检测得出,与对照组相比,抗霉素A组细胞琥珀酸脱氢酶相对活性显著下降(P<0.0001)、乳酸脱氢酶相对活性增强(P<0.01)、细胞迁移能力明显下降(P<0.0001),三溴丙酮酸组细胞琥珀酸脱氢酶相对活性显著下降(P<0.0001)、乳酸脱氢酶相对活性无明显变化(P>0.05)、细胞迁移能力下降(P<0.001).线粒体通过氧化磷酸化为肺腺癌细胞的迁移提供能量,促进了肺腺癌细胞的迁移. A549 lung cancer cells were cultured by conventional method,antimycin A(mitochondrial function inhibitors),three bromine pyruvate(glycolysis functional inhibitor)and sodium pyruvate were added to them,and the optimal drug concentrations were determined by the MTT method,A549 lung cancer cells were treated with this optimal drug concentrations.The function of mitochondrial oxidative phosphorylation was detected with histochemical and scratch method in the migration of A549 lung cancer cells.The following results were obtained:the optimal concentrations of antimycin A,three bromine pyruvate and sodium pyruvate tested by MTT method was 150μmol/L,200μmol/L and 0.02 mol/L respectively.The results of histochemical and scratch test showed that lactate dehydrogenase activity of antimycin A group was significantly higher than that of control group(P<0.01),succinate dehydrogenase activity and migration distance of antimycin A group were significantly lower compared with control group(P<0.0001),the succinate dehydrogenase activity of three bromine pyruvate group was significantly lower than that of control group(P<0.0001),there was no significant difference(P>0.05)between the lactate dehydrogenase activity of three bromine pyruvate group and control group,and the cell migration distance of three bromine pyruvate group was lower compared with control group(P<0.001).Mitochondrial oxidative phosphorylation not only provided enough energy for lung adenocarcinoma cells migration,but also promoted the migration of lung adenocarcinoma cells.
