双相情感障碍患者血浆lnc RNA BDNF-AS和GOMAFU的检测水平及意义 被引量:9

The Detection Levels and Significances of LNC RNA BDNF-AS and GOMAFU in Plasma of Patients with Bipolar Affective Disorder

摘要 目的检测双相情感障碍患者血浆脑源性神经营养因子反义长链非编码lnc RNA(brainderived neurotrophic factor-antisense long-chain non-coding RNA,lnc RNA BDNF-AS)和GOMAFU[一种长链非编码RNA,又称为MIAT(myocardial infarction associated transcript,心肌梗死相关转录本)]水平,并探讨其临床意义。方法选取2016年9月~2018年9月本院收治的112例双相情感障碍患者(躁狂组58例,抑郁组54例)、46例精神分裂症患者及45例健康体检者为研究对象,采用qRT-PCR测定受试者血浆lnc RNA BDNF-AS、脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)和GOMAFU表达水平,分析BDNF-AS、GOMAFU诊断价值并探究两者与症状相关性。结果精神分裂组、双相情感障碍组血浆BDNF-AS水平较对照组显著升高(P<0.05),且双相情感障碍组较精神分裂组高,BDNF与GOMAFU水平较对照组显著降低,且双相情感障碍组较精神分裂组低(P<0.05);血浆BDNF-AS、GOMAFU水平诊断双相情感障碍的曲线下面积(area under the curve,AUC)分别为0.843、0.736,分界点分别为6.75、0.68,敏感度分别为79.83%、95.56%,特异度分别为79.76%、57.26%;狂躁组和抑郁组血浆BDNF-AS水平较对照组显著升高,且狂躁组较抑郁组高,而BDNF与GOMAFU水平较对照组显著降低,且狂躁组较抑郁组低(P<0.05);狂躁组、抑郁组血浆BDNF-AS和GOMAFU水平与杨氏躁狂评定量表(Young manic rating scale,YMRS)及汉密尔顿抑郁量表(Hamilton rating scale for depression,HAMD)评分相关性均无统计学意义(P>0.05)。结论双相情感障碍患者血浆lnc RNA BDNF-AS水平上调,GOMAFU水平下调,二者可能作为临床诊断潜在参考指标。 Objective To detect the levels of LNC RNA BDNF-AS and GOMAFU in plasma of patients with bipolar affective disorder and to explore their clinical significances. Methods From September 2016 to September 2018,112 patients with bipolar affective disorder(58 in manic group,54 in depression group),46 patients with schizophrenia,and 45 healthy physical examiners were selected as subjects,the levels of plasma LNC RNA,BDNF-AS,brainderived neurotrophic factor(BDNF),and GOMAFU were measured by qRT-PCR,the diagnostic value of BDNF-AS and GOMAFU were analyzed,and explored the correlation between them and symptoms. Results The plasma levels of BDNF-AS in schizophrenia group and bipolar affective disorder group were significantly higher than those in control group(P<0.05). The levels of BDNF and GOMAFU in bipolar affective disorder group were significantly lower than those in control group,and the levels of BDNF and GOMAFU in bipolar affective disorder group were lower than those in schizophrenia group(P<0.05). The areas under the curve(AUC)of plasma BDNF-AS and GOMAFU levels in the diagnosis of bipolar affective disorder were 0.843 and 0.736,respectively. The demarcation points were 6.75 and 0.68,the sensitivity was 79.83% and 95.56%,the specificity was 79.76% and 57.26%,respectively;the plasma levels of BDNF-AS in manic group and depression group were significantly higher than those in control group,and the levels of BDNF and GOMAFU in manic group were significantly lower than those in depression group(P< 0.05). There was no significant correlation between plasma BDNF-AS and GOMAFU levels and Young manic rating scale(YMRS)and Hamilton rating scale for depression(HAMD)in manic and depressive groups(P> 0.05). Conclusion The level of plasma LNC RNA BDNF-AS in patients with bipolar affective disorder is up-regulated,GOMAFU level is downregulated,both of them may serve as potential indicators for clinical diagnosis.
