摘要
为了得到大量欧洲缬草无菌苗和不定根,本研究以欧洲缬草种子为实验材料,研究不同消毒方法对种子发芽的影响;不同培养基种类对无菌苗生长的影响;以及不同生长素对无菌苗根段不定根诱导的影响。结果发现:种子发芽的最佳消毒方法为75%的酒精溶液浸泡消毒30 s,4%的NaClO溶液浸泡消毒10 min,无菌水冲洗3~5次;White培养基的种子发芽率、无菌苗茎叶和根长都较为理想,是欧洲缬草利用种子获得无菌苗的理想培养基。另外,WPM培养基对种子发芽和无菌苗茎叶生长具有明显的促进作用,B_5培养基则有利于无菌苗根系生长;MS+0.1 mg/L IBA是根段为外植体诱导不定根的理想培养基,不定根诱导率高达100%。本研究为欧洲缬草无菌苗和不定根的诱导繁殖提供基础依据。
In order to obtain a large number of Valeriana officinalis sterile seedlings and adventitious roots,the seeds of V.officinalis have been used to study the effects of different disinfection methods on seed germination,the effects of different media types on the growth of sterile seedlings,and the effects of different auxins on adventitious root induction in sterile seedlings roots segment.The results showed that the best disinfection method for seed germination was 75%alcohol solution soaked for 30 s,4%NaClO solution soaked for 10 min,and sterile water washed for 3~5 times.Besides,white medium was an ideal medium of seed germination,and the growth of sterile seedling stem,roots.It was a better medium to obtain sterile seedlings.In addition,WPM medium had obvious promoting effect on seed germination and stem growth of sterile seedling,and B5 medium was beneficial to root growth of sterile seedlings.MS+0.1 mg/L IBA was the ideal medium for sterile seedlings roots segment to induce adventitious roots,and the adventitious root induction rate was 100%.This research provides a technical basis for the breeding of Valeriana officinalis weeds and the mass reproduction of adventitious roots.
