摘要
目的用全基因组芯片筛选出在类风湿性关节炎发生发展中差异表达热休克蛋白90(HSP90)客户蛋白的基因。方法按照体重将SD大鼠随机分为2组:正常对照组和模型组,每组3只。模型组大鼠左后足皮下注射弗氏完全佐剂0.1 mL制备关节炎模型,正常对照组予以等量0.9%NaCl。造模后第21天,深度麻醉下取2组大鼠滑膜组织,用Array star大鼠全基因组芯片来检测差异表达的信使核糖核酸(mRNA),再根据已知的942个HSP90的客户蛋白进一步筛选对应的mRNA。结果筛选出85个与佐剂性关节炎发生发展有关的差异表达HSP90客户蛋白的mRNA,其中上调53个,倍数最高的是白细胞介素-1β(IL-1β);下调32个,倍数最高的是莫霍克同源框(MKX)。结论在HSP90的客户蛋白中,IL-1βmRNA等53个上调的mRNA和MKX等32个下调的mRNA与佐剂性关节炎发生发展密切相关。
Objective To screen out the mRNA of differential expression associated with heat shock protein(HSP90)client protein in the development of rheumatoid arthritis(RA)by using genome-wide chips.Methods SD rats were randomly divided into two groups according to weight:Normal control group and model group,with 3 rats in each group.The rats in the model group were subcutaneously injected with 0.1 mL Freund’s complete adjuvant to prepare an arthritis model.The normal control group was given an equal amount of saline.On day 21,the synovial tissues of rats in two groups were taken under deep anesthesia and the Array star rat whole genome chip was used to detect differentially expressed messenger ribonucleic acid(mRNA).Screen out the related mRNA with the known 942 HSP90 client proteins.Results Eighty-five differentially expressed HSP90 client protein mRNAs related to the occurrence and development of adjuvant arthritis were screened out.Among them,53 were up-regulated and the highest fold was interleukin-1β(IL-1β);32 were down-regulated and the highest fold was Mohawk homeobox(MKX).Conclusion Among HSP90 client proteins,53 up-regulated mRNAs such as IL-1βmRNA and 32 down-regulated mRNAs such as MKX are closely related to the development of adjuvant arthritis.
