摘要
目的:探讨痹证宁对风寒湿痹型类风湿关节炎JAK-STAT信号通路的影响及作用机制。方法:将70只Wistar大鼠按照随机数字表法分为空白对照组,模型对照组,痹证宁低、中、高剂量组(9,18,36 g·kg-1),甲氨蝶呤组(1 mg·kg-1)和中西医结合组(甲氨蝶呤1 mg·kg-1+痹证宁18 g·kg-1),每组10只。除空白对照组外,其余各组大鼠模拟风寒湿环境,采用Ⅱ型胶原诱导方法制备风寒湿痹型类风湿关节炎大鼠模型,各组大鼠给予相应药物干预。28 d后,观察大鼠关节肿胀度、关节滑膜组织白细胞介素-6(IL-6)含量、PIAS3、P-STAT3蛋白和JAK3、STAT3 mRNA表达情况。结果:痹证宁可显著降低风寒湿痹型类风湿关节炎模型大鼠的关节肿胀度,降低IL-6含量,抑制P-STAT3蛋白、JAK3和STAT3 mRNA过表达,增进PIAS3蛋白表达。结论:痹证宁有抗炎、免疫调节作用,其作用机制可能与抑制JAK-STAT信号通路的过度激活有关。
Objective:To explore the effect of Bizheng Ning(痹证宁)on JAK-STAT signal pathwayin rheumatoid arthritis of wind-cold-dampness bi type and its mechanism.Methods:Seventy Wistar rats were randomly divided into a blank control group,a model control group,low,medium and high dose Bizheng Ning groups(9,18,36 g·kg-1),a methotrexate group(1 mg·kg-1)and a combination group(1 mg·kg-1 of methotrexate+18 mg·kg-1 of Bizheng Ning)treated with Chinese and Western medicine.There were10 rats in each group.Except the blank control group,the other groups of rats were used to establish rheumatoid arthritis models of wind-cold-dampness bi type by the method of typeⅡcollagen induction.The rats in each group were given corresponding medicine intervention.After 28 days,the degree of joint swelling,the content of IL-6 in synovium tissue,PIAS3,the expressions of P-STAT3 protein,JAK3 and STAT3 mRNA were observed.Results:Bizheng Ning can significantly reduce the joint swelling and IL-6 content,inhibit the expressions of P-STAT3 protein,JAK3 and STAT3 mRNA,and increase the expression of PIAS3 protein.Conclusion:Bizheng Ning has anti-inflammatory and immunomodulatory effects,whose mechanism may be related to inhibiting the over activation of JAK-STAT signal pathway.
