葛根素对妊娠高血压模型大鼠血管活性物质及胎盘组织的影响 被引量:4

Effects of puerarin on vasoactive substances and placental tissues of rats with pregnancy induced hypertension

摘要 目的研究葛根素对妊娠高血压(PIH)模型大鼠血管活性物质及胎盘组织可溶性血管内皮生长因子受体-1(sFLT-1)和胎盘生长因子(PLGF)表达的影响。方法将45只妊娠第12天的SD大鼠随机分为对照组(n=15),模型组(n=15)和实验组(n=15)。模型组和实验组大鼠用连续7 d灌胃亚硝基左旋精氨酸甲酯(80 mg·kg^-1·d^-1)的方法建立PIH大鼠模型。造模后,实验组大鼠给予葛根素(50 mg·kg^-1·d^-1),灌胃,对照组与模型组灌胃等量生理盐水,连续干预8周。干预结束后,用酶联免疫吸附(ELISA)法检测血清精氨酸加压素(AVP)、内皮素(ET)、血管紧张素Ⅱ(AngⅡ)水平;用蛋白质印迹(WB)法检测胎盘组织sFLT-1、PLGF蛋白表达水平。结果对照组、模型组、实验组大鼠血清AVP水平分别为(21.37±1.25),(39.23±2.62),(28.12±1.46)ng·L^-1;ET水平分别为(0.76±0.01),(1.65±0.11),(1.12±0.06)ng·mL^-1;AngⅡ水平分别为(218.73±13.42),(384.12±75.56),(261.72±43.69)ng·L^-1;胎盘组织sFLT-1蛋白相对表达量分别为0.12±0.05,0.96±0.04,0.47±0.11;PLGF蛋白相对表达量分别为0.95±0.05,0.14±0.07,0.48±0.13。与对照组比较,模型组大鼠血清AVP、ET、AngⅡ水平及胎盘组织sFLT-1蛋白相对表达量均升高,实验组显著低于模型组;PLGF蛋白相对表达量降低,且实验组高于模型组(均P<0.05)。结论葛根素可抑制PIH大鼠血管活性物质水平,改善肾动脉病变,同时下调胎盘组织sFLT-1蛋白表达,上调PLGF蛋白表达,进而达到治疗PIH的目的。 Objective To investigate the effects of puerarin on vasoactive substances and expression of soluble vascular endothelial growth factor receptor-1(sFLT-1)and placental growth factor(PLGF)in placental tissues of rats with pregnancy induced hypertension(PIH).Methods Forty-five SD rats on the 12th day of pregnancy were randomly divided into control group(n=15),model group(n=15)and test group(n=15).PIH rat models were established by gavage of nitroso-L-arginine methyl ester(80 mg·kg^-1·d^-1)for 7 days in model and test groups.After modeling,rats in test group were gavage with puerarin(50 mg·kg^-1·d^-1),control group and model group were gavage with the equal amount of normal saline for 8 weeks.After intervention,the levels of serum arginine vasopressin(AVP),endothelin(ET),and angiotensinⅡ(AngⅡ)were measured by enzyme-linked immunosorbent assay(ELISA);Western blotting(Wb)method was used to detect the expression levels of sFLT-1 and PLGF in placental tissues.Results The serum AVP levels in control,model and test groups were(21.37±1.25),(39.23±2.62)and(28.12±1.46)ng·L^-1;ET levels were(0.76±0.01),(1.65±0.11)and(1.12±0.06)ng·mL^-1;AngⅡlevels were(218.73±13.42),(384.12±75.56)and(261.72±43.69)ng·L^-1;the relative expression of sFLT-1 protein in placenta tissues were 0.12±0.05,0.96±0.04,0.47±0.11;the relative expression of PLGF protein were 0.95±0.05,0.14±0.07,0.48±0.13.Compared with control group,the levels of serum AVP,ET,AngⅡand the relative expression of sFLT-1 protein in placenta tissues of model group were increased,and test group were significantly lower than those in model group;the relative expression of PLGF protein in model group was decreased,and test group was higher than model group(all P<0.05).Conclusion Puerarin can inhibit the levels of vasoactive substances in PIH rats,improve renal artery disease,and at the same time down-regulate the expression of sFLT-1 protein in the placental tissues and up-regulate the expression of PLGF protein,thereby achieving the purpose of treating P
