摘要
目的调查河北省廊坊地区汉族女性5,10-亚甲基四氢叶酸还原酶(MTHFR)C677T、A1298C位点基因多态性分布特征,研究分析血清同型半胱氨酸(HCY)与遗传因素的相关性,检测该地区孕期汉族女性血清同型半胱氨酸(HCY)水平,分析孕期不同阶段HCY水平变化。方法以该地区922位汉族女性为研究对象,检测其MTHFR C677T、A1298C基因位点多态性,与我国其他南北各地区既有数据对比,采用统计学方法分析基因的多态性分布特征;检测其孕早、孕中、孕后期不同阶段HCY水平,同时测定其中192位女性产后42 d HCY水平,分析该生化指标与遗传因素的相关性。结果本地区孕期汉族女性MTHFR 677TT基因型频率为33.5%,MTHFE C677T三种基因型对孕期血清HCY水平差异无统计学意义(P>0.05);192名女性孕期与产后42 d血清HCY水平比较,差异有统计学意义(P<0.05)。结论本地区汉族女性MTHFR C677T基因频率分布特征有其自身的特点;基因位点的频率变化对孕期血清HCY水平无显著性影响,TT型对产后42 d血清HCY水平有显著性影响。
Objective To investigate the distribution characteristics of 5,10 methylenetetrahydrofolate reductase(MTHFR)C677T,A1298C gene polymorphisms,and to explore the correlation between the serum homocysteine(HCY)and genetic factors in Han women in Langfang area,and to investigate the levels of HCY and the changes of HCY in pregnant women at different stages of pregnancy.Methods A total of 922 Han women in this area were enrolled as research objects.The levels of HCY were detected and compared amon the pregnant women at early period,middle period,late period of pregnancy,of whom,the levels of HCY in 192 puerpera at 42 days after childbirth were detected.In addition the gene polymorphisms of MTHFR C677T was detected by using fluorescence quantitative PCR.And the correlation between HCY and MTHFR C677T was analyzed.Results The frequency of the MTHFR 677TT genotypes in Langfang area was 35.8%.There were no significant differences in HCY levels among early pregnancy,middle pregnancy and late pregnancy(P>0.05).There were statistically significant differences in HCY levels in the 192 pregnant women between pregnant period and at 42 days after childborth(P<0.05).Conclusion The gene frequency distribution of MTHFR in the Han women in Langfang area has its own characteristics,and the changes of gene locus have no significant effects on HCY levels during pregnancy,however,TT type has significant effects on the HCY levels of puerpera at 42 days after childbirth.
