Simultaneous determination of eight constituents in Fengliaoxing Fengshi Dieda Wine by HPLC

摘要 目的建立HPLC法同时测定冯了性风湿跌打药酒中新绿原酸、东莨菪苷、绿原酸、隐绿原酸、东莨菪内酯、异绿原酸B、异绿原酸A、异绿原酸C的含量,为冯了性风湿跌打药酒的质量控制提供参考。方法采用Agilent 5 TC-C18色谱柱(250 mm×4.6 mm,5μm),乙腈-0.1%磷酸作流动相,梯度洗脱,流速1.0 mL·min-1,柱温25℃,检测波长345 nm。结果新绿原酸、东莨菪苷、绿原酸、隐绿原酸、东莨菪内酯、异绿原酸B、异绿原酸A、异绿原酸C分离度良好且分别在0.86~86.00、1.28~128.00、1.10~110.00、0.72~72.00、0.86~86.00、2.18~218.00、1.34~134.00、1.76~176.00μg·mL-1范围内呈良好的线性关系(r≥0.9998);8种成分的平均加样回收率在97.78%~101.45%之间,RSD在0.95%~1.90%之间。结论该方法简便、准确,分离效果好,重复性好,可用于冯了性风湿跌打药酒的质量评价。 Objective An accurate and reliable HPLC method was developed to simultaneously determine the contents of eight active components(including neochlorogenic acid, scopolin, chlorogenic acid, cryptochlorogenic acid, scopoletin, isochlorogenic acid B, isochlorogenic acid A and isochlorogenic acid C) in Fengliaoxing Fengshi Dieda Wine, to provide references for its quality control. Methods Eight active components in the samples were all well separated by gradual elution with acetonitrile-0.1% phosphate acid solution. The chromatographic conditions: Agilent 5 TC-C18 column, the flow rate of the mobile phase was 1.0 mL·min-1 with the column temperature of 25℃, and the detection wavelength was set at 345 nm. Results Neochlorogenic acid, scopolin, chlorogenic acid, cryptochlorogenic acid, scopoletin, isochlorogenic acid B, isochlorogenic acid A and isochlorogenic acid C were well separated and showed good linear relationship within the ranges of 0.86~86.00,1.28~128.00,1.10~110.00,0.72~72.00,0.86~86.00,2.18~218.00,1.34~134.00,1.76~176.00 μg·mL-1(r≥0.9998), whose average recoveries were 97.78%~101.45% with the RSDs of 0.95%~1.61%. Conclusion This sample and accurate method can be used for the quality control of Fengliaoxing Fengshi Dieda Wine.
