甜菊糖苷干预对高脂血T2DM大鼠生化指标的影响 被引量:3

摘要 目的:探讨甜菊糖苷干预对高脂血T2DM型大鼠的降糖降脂作用。方法:高脂高糖饮食4周后,链脲佐菌素(50mg/kg)腹腔注射至Wistar(SPF)雄性大鼠60只,建高脂血T2DM模型。随机分为模型组,甜菊糖苷组(低、中、高剂量),二甲双胍组,另设正常对照组。甜菊糖苷组(低、中、高剂量)分别灌胃甜菊糖苷50mg(kg/d)、100mg(kg/d)、200mg(kg/d),二甲双胍组灌胃50mg(kg/d),持续干预28d。观察对空腹血糖、糖耐量、总胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白、胰岛素水平、抗氧化因子SOD、MAD活性等的影响。结果:长期中、高剂量甜菊糖苷干预可显著降低高脂血型T2DM大鼠空腹血糖、总胆固醇、甘油三脂、低密度脂蛋白、MAD活性(P<0.01),明显提高大鼠葡萄糖耐受性、刺激胰岛素分泌、增加高密度脂蛋白含量和SOD活性。结论:长期高剂量甜菊糖苷干预,可有效调节高脂血T2DM大鼠的高血糖并发高脂血症,促进胰岛素分泌,增加糖耐量,增强抗氧化能力,抑制动物体内脂质过氧化,这对降糖降脂均具有一定的作用,并呈现剂量依赖性。由此可推断,甜菊糖苷干预对高脂血型大鼠T2DM及其心血管并发症有一定辅助疗效,甜菊糖苷可以作为T2DM型患者的代糖品。 Objective:To study the stevia glycoside intervention to hyperlipidemiaⅡdiabetes model rats blood glucose and blood lipid.Methods:After 4 weeks of high-fat diet,streptozotocin(40 mg/kg)was intraperitoneally injected into SD rats to establish an animal model of hyperlipidemia type 2 diabetes.Rats were randomly divided into model group,stevia glycoside group(low,medium and high dose),metformin group,and blank group.Stevia glycoside groups(low,middle and high dose)were administrated by oral gavage of stevia glycoside at 50 mg(kg/d),100 mg(kg/d),200 mg(kg/d),metformin group were administrated by oral gavage of metformin at 50 mg(kg/d)for total of 28 d.Blood sugar,glucose tolerance,total cholesterol,triglycerides,high-density lipoprotein cholesterol(hdl-c),low density lipoprotein,insulin levels,anti-oxidation factor of SOD,MAD were all tested.Results:Middle and high dose stevia glycoside can obviously reduce the level of fasting blood glucose(P<0.01),total cholesterol,triglyceride,low density lipoprotein cholesterol(hdl-c),MAD activity,and greatly increase glucose tolerance,insulin,high-density lipoprotein cholesterol(hdl-c),SOD content.Conclusion:High dose of stevia sugar can effectively adjust the hyperlipidemiaⅡdiabetes model rats of hyperglycemia and hyperlipidemia,promote insulin secretion,increase the glucose tolerance,enhance SOD activity and reduce the activity of MAD,and inhibit lipid peroxidation inhibition.It is helpful in reducing blood sugar and blood lipid,and is a dose dependent.Therefore,stevia sugar showed certain effects for hyperlipidemiaⅡdiabetes model rats.Stevia glycoside can be used as a sugar substitute for T2 DM patients.
