摘要
目的探讨甘草酸二铵联合丹参注射液对四氯化碳(CCl4)诱导急性肝损伤小鼠保护作用的协同增效情况。方法将50只雄性KM小鼠随机分为空白组、模型组、甘草酸二胺注射液组(70 mg·kg^-1)、甘草酸二胺(70 mg·kg^-1)+丹参注射液(0.5 g·kg^-1)组、多烯磷脂酰胆碱注射液组(140 mg·kg^-1)。尾静脉注射给药,每日1次,连续3 d;按0.01 mL·g-1腹腔注射0.4%的CCl4橄榄油溶液诱导小鼠急性肝损伤模型。检测血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)的含量;采用染料法荧光定量PCR检测肝脏组织中白细胞介素1β(IL^-1β)及肿瘤坏死因子α(TNF-α)mRNA表达水平;采用HE染色法观察肝脏组织病理变化情况。结果与空白组比较,模型组小鼠血清ALT、AST水平均明显升高(P<0.01),肝脏组织IL^-1β、TNF-αmRNA表达均明显上调(P<0.01)。与模型组比较,各给药组小鼠血清的ALT、AST水平均明显降低(P<0.01),肝脏组织IL^-1β、TNF-αmRNA表达均明显下调(P<0.01)。与甘草酸二胺注射液组比较,甘草酸二胺+丹参注射液组的血清ALT、AST水平均明显降低(P<0.01),肝脏组织IL^-1β、TNF-αmRNA表达明显下调(P<0.01)。病理检查结果显示,各给药组均能不同程度减轻肝脏组织病变。结论甘草酸二胺注射液与丹参注射液联合用药对CCl4诱导的急性肝损伤小鼠的保护具有协同增效作用,可能与其增强对炎症因子的调控有关。
Objective To study the synergistic effect of diammonium glycyrrhizinate injection and salvia injection on the protection of acute liver injury induced by CCl4 in mice.Methods Fifty KM mice were randomly divided into blank group,model group,diammonium glycyrrhizinate injection group(70 mg·kg^-1),diammonium glycyrrhizinate injection(70 mg·kg^-1)+salvia injection group(0.5 g·kg^-1),polyene phosphatidylcholine injection group(140 mg·kg^-1).The mouse model of acute liver injury was induced by intraperitoneal injection with 0.4%CCl4 olive oil solution.After injection of drugs for 3 days,blood sample was collected,the animals were sacrificed and liver tissue samples were taken.Serum contents of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)were detected.Fluorescence quantitative PCR with dye method was applied to detect mRNA levels of interleukin-1β(IL^-1β)and tumor necrosis factor-α(TNF-α)in liver tissue;and HE staining was used to observe the pathological changes of liver tissue.Results Compared with the blank group,the ALT and AST levels of the model group were significantly increased(P<0.01).The relative amounts of IL^-1βmRNA and TNF-αmRNA were significantly increased(P<0.01).Compared with the model group,the serum levels of ALT and AST indrug treated groups were significantly decreased(P<0.01);the relative amounts of IL^-1βmRNA and TNF-αmRNA were significantly reduced(P<0.01).Compared with the diammonium glycyrrhizinate injection group,the serum levels of ALT and AST in the diammonium glycyrrhizinate injection+salvia injection group were significantly reduced(P<0.01);the relative amounts of IL^-1βmRNA and TNF-αmRNA were significantly reduced(P<0.01).Pathological examination show that polyene phosphatidylcholine injection,diammonium glycyrrhizinate injection,diammonium glycyrrhizinate injection combined with salvia injection can significantly reduce liver tissue lesions.Conclusion Diammonium glycyrrhizinate injection combined with salvia injection synergistically protect the livers of
