摘要
目的建立一测多评法同时测定华佗延寿酒中原儿茶酸、原儿茶醛、儿茶素、表儿茶素、薯蓣皂苷元、茅术醇、β-桉叶醇和苍术素8种主要成分。方法采用Zorbax Extend C18色谱柱,以甲醇-乙腈(体积比1:1)与体积分数0.5%冰醋酸溶液为流动相,梯度洗脱,检测波长为280 nm(原儿茶酸、原儿茶醛、儿茶素、表儿茶素)、203 nm(薯蓣皂苷元、茅术醇、β-桉叶醇)和340 nm(苍术素),以表儿茶素为内标物,计算原儿茶酸、原儿茶醛、儿茶素、薯蓣皂苷元、茅术醇、β-桉叶醇和苍术素的相对校正因子,测定其含有量。结果原儿茶酸、原儿茶醛、儿茶素、表儿茶素、薯蓣皂苷元、茅术醇、β-桉叶醇和苍术素分别在0.91~22.75、2.99~74.75、0.78~19.50、2.21~55.25、1.39~34.75、3.18~79.50、3.92~98.00和2.69~67.25 mg·L-1内线性关系良好,平均加样回收率分别为97.31%、98.96%、96.98%、99.33%、97.97%、98.47%、100.01%和98.83%,RSD分别为1.20%、1.06%、1.46%、0.80%、1.38%、1.23%、0.63%和1.13%,结论一测多评法和外标法测定结果无显著性差异。
Objective A quantitative analysis of multi-components by single-marker(QAMS)method was established for the determination of protocatechuic acid,protocatechuic aldehyde,catechin,epicatechin,diosgenin,hinesol,β-eudesmol and atractylodin in Huatuo Yanshou Jiu.Methods A Zorbax Extend C18 column(250 mm×4.6 mm,5μm)was methanol-acetonitrile(1∶1)and 0.5%glacial acetic acid solution by gradient elution.Compounds protocatechuic acid,protocatechuic aldehyde,catechin and epicatechin were detected at 280 nm,compounds diosgenin,hinesol andβ-eudesmol were detected at 203 nm,and compounds atractylodin was detected at 340 nm.With epicatechin as an internal standard,the relative correction factors of protocatechuic acid,protocatechuic aldehyde,catechin,diosgenin,hinesol,β-eudesmol and atractylodin were calculated,from which the content determination was made.Results The linear ranges for the above eight components were 0.91-22.75,2.99-74.75,0.78-19.50,2.21-55.25,1.39-34.75,3.18-79.50,3.92-98.00,2.69-67.25 mg·L-1,respectively.The average recoveries were 97.31%(1.20%),98.96%(1.06%),96.98%(1.46%),99.33%(0.80%),97.97%(1.38%),98.47%(1.23%),100.01%(0.63%)and 98.83%(1.13%).RSDs were 1.20%,1.06%,1.46%,0.80%,1.38%,1.23%,0.63%and 1.13%.Conclusion The results showed that there were no significant differences between QAMS method and the external standard method.
