新止骨增生丸对膝骨关节炎软骨细胞JAK-STAT信号通路下JAK2、P-JAK2表达及SOCS1负反馈影响 被引量:6

摘要 目的观察新止骨增生丸含药血清对软骨细胞JAK-STAT信号通路相关调节因子的影响,探讨新止骨增生丸对骨关节炎的作用机制。方法C28/I2细胞系建立体外软骨细胞退变模型,分别采用空白血清、新止骨增生丸含药血清、抑制剂血清、抑制剂+含药血清干预24 h后,收集软骨细胞,采用ELISA检测软骨细胞分泌肿瘤细胞坏死因子(TNF-α)和白细胞介素(IL-6)能力的影响,Westernblot法检测JAK2、SOCS1蛋白表达水平变化,Westernblot法检测各组软骨细胞中JAK2的磷酸化水平。结果与模型组相比,ELISA结果表明新止骨增生丸含药血清可有效降低炎性因子TNF-α和IL-6含量(P<0.05);Westernblot结果与ELISA结果趋势一致,即新止骨增生丸含药血清能抑制JAK2蛋白表达(P<0.05)。SOCS1负反馈调节蛋白表达升高。结论新止骨增生丸可通过调控JAK-STAT信号通路,抑制骨关节炎炎症反应,从而起到治疗骨关节炎作用。 Objective To observe the effects of New Zhigu Zengsheng Pill-containing serum on the regulatory factors of JAK-STAT1 signaling pathway in chondrocytes,and to explore the mechanism of New Zhigu Zengsheng Pill on osteoarthritis.Methods The C28/I2 cell line was used to establish an in vitro chondrocyte degeneration model.After intervention with blank serum,New Zhigu Zengsheng Pill drugcontaining serum,inhibitor serum,inhibitor+drug-containing serum for 24 hours,chondrocytes were collected and detected by ELISA.The effect of chondrocytes on the ability to secrete TNF-αand IL-6.Westernblot method was used to detect the expression of JAK2 and SOCS1 protein.Westernblot method was used to detect the phosphorylation level of JAK2 in chondrocytes of each group.Results Compared with the model group,the ELISA results showed that the New Zhigu Zengsheng Pill containing medicated serum can effectively reduce the levels of inflammatory factors TNF-αand IL-6(P<0.05);the Westernblot results are consistent with the ELISA results trend,that is,New Zhigu Zengsheng Pill can inhibit the expression of JAK2 protein in serum(P<0.05).SOCS1 negative feedback regulatory protein expression was increased.Conclusion New Zhigu Zengsheng Pill can treat osteoarthritis by regulating JAK-STAT1 signal pathway and inhibiting inflammatory reaction of osteoarthritis.
