两种基于空间位阻、用于识别甘油糖脂的专一性蛋白的有效合成方法 被引量:3

摘要 结合细胞表面脂质膜的结构信息,设计以甘油为骨架、以α-甘露糖为头基、寡聚乙二醇(OEG)为间链链接的饱和双亲水基团与疏水脂链的甘油双糖头基糖脂结构,采用三氟甲磺酸银(AgOTf)和四丁基溴化铵(TBAB)作为空间位阻效应相转移的催化剂有效合成了甘油骨架连接两亲间链OEG糖脂(BEMP)和不含OEG间链糖脂(BMP)以及其前驱体双十六烷基醚寡聚乙二醇(BEG)和2-十六烷基醚甘油二醇(BHG),以较少合成步骤和较简单方法实现了BEMP和BMP的产率达到70%以上,为专一性识别小分子糖脂的蛋白高产率合成提供了新方法。 Based on the structure information of lipid membrane on the cell surface,this paper presents the design and synthesis of two kinds of amphiphile glycolipids BEMP and BMP,and their protein-repelling lipids BEG and BHG.BEMP and BMP are similar with structure of glycolipid with glycerol as the skeleton,α-mannose as the head group,oligo polyethylene glycol(OEG)as the chain with glycerol head group disaccharide backbone.This structure consists of three parts:α-mannose for headgroups,oligoethylene glycol(OEG)for spacer chain and glycerol backbone to connect two hydrophilic groups with saturated aliphatic hydrophobic chain.Silver trifluorome than esulfonate(AgOTf)and tetrabutylammonium bromide(TBAB)are used as the catalysts for steric hindrance phase transfer to synthesize glycolipids and the yield of BEMP and BMP is over 70%with less steps and simple methods,which provides a new and high yield synthetic method of proteins for specific recognition of small molecular glycolipid.
