摘要
[目的]本文旨在研究酶促反应合成的18种含有不同脂肪酸链数量及碳链长度(C 8∶0—C 18∶3)的半乳糖基甘油糖脂对5株蜡样芽胞杆菌(Bacillus cereus)的抑菌活性,并初步探讨其活性构效关系。[方法]通过测定半乳糖基甘油糖脂对蜡样芽胞杆菌的抑菌圈直径、最小抑菌浓度(MIC)、最小杀菌浓度(MBC)以及抑菌率来研究半乳糖基甘油糖脂的抑菌活性及其构效关系。[结果]本试验共合成9种半乳糖基甘油单酯和9种酰化半乳糖基甘油单酯。半乳糖基肉豆蔻酸甘油单酯(MMGG)的抑菌效果最好,对5株蜡样芽胞杆菌的抑菌圈直径均为16~20 mm,MIC值和MBC值均为0.039 mg·mL^(-1),且抑菌率均在99%以上;半乳糖基月桂酸甘油单酯等其余含有饱和中链脂肪酸(C 8∶0—C 14∶0)的半乳糖基甘油糖脂以及含有不饱和脂肪酸的半乳糖基甘油糖脂的抑菌效果次之;在含有不饱和脂肪酸的半乳糖基甘油糖脂中,半乳糖基亚麻酸甘油单酯的抑菌效果最好,而含有饱和长链脂肪酸(C 16∶0、C 18∶0)的半乳糖基甘油糖脂抑菌效果较差。半乳糖基甘油糖脂的抑菌活性与其所含脂肪酸链的不饱和度呈极显著负相关,与其所含脂肪酸链的长度存在截断效应,而不是简单的线性关系。[结论]脂肪酸链的链长、数量以及不饱和度都会影响半乳糖基甘油糖脂的抑菌活性。
[Objectives]The aim of this study was to investigate the inhibitory activities of 18 kinds of galactosylglycerides with different number and length(C 8∶0-C 18∶3)of fatty acyl chains against Bacillus cereus,and the structure-activity relationship was preliminarily discussed.[Methods]The antibacterial activity and structure-activity relationship of galactosylglycerides against B.cereus were investigated by measuring the diameter of inhibition zone,minimum inhibitory concentration(MIC),minimum bactericidal concentration(MBC)and inhibition rate.[Results]9 kinds of galactosylglycerides bearing one acyl chain and 9 kinds of galactosylglycerides bearing two acyl chains were synthesized.1(6′)-monomyristoyl-3-O-β-D-galactopyranosyl-sn-glycerol(MMGG)had the best antibacterial effect.MIC and MBC of MMGG against 5 strains of B.cereus were 0.039 mg·mL^(-1).The inhibition diameter of MMGG was 16-20 mm and the inhibition rate of MMGG was above 99%.The following were other galactosylglycerides containing saturated medium fatty acyl chains(C 8∶0-C 14∶0)and unsaturated fatty acyl chains.Among galactosylglycerides with unsaturated fatty acyl chains,1(6′)-monolinolenoyl-3-O-β-D-galact-opyranosyl-sn-glycerol had the best antibacterial effect.The antibacterial effect of galactosylglycerides containing saturated long fatty acyl chains was poor.The antibacterial activity of galactosylglycerides was negatively correlated with unsaturation of fatty acyl chains,and there was a cut-off effect on the length of fatty acyl chains,rather than a simple linear relationship.[Conclusions]The fatty acyl chain length and number,and the degree of saturation played an important role in the antibacterial activity of galactosylglycerides.
