蒙药蓝刺头抗PMOP骨组织计量学研究 被引量:9

摘要 目的:为蒙医药基于“补暖补精-清镇赫依-固骨质壮骨”经典理论防治绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)提供实验依据,为蒙医药现代化提供思路。方法:将4月龄SD系SPF级健康雌性未孕大鼠49只随机分为假手术组、模型组、淫羊藿组、辅酶Q10组及蒙药蓝刺头高、中、低剂量组。采用去势方法制造PMOP模型,阴道上皮角化实验方法验证造模是否成功。术后饲养90天,模型组、假手术组生理盐水灌胃,其余各组予相应药物灌胃处死大鼠,剔取右侧完整股骨,4%多聚甲醛溶液固定保存。检测右侧股骨离体骨骨密度(bone mineral density,BMD),包括股骨近端BMD及股骨总BMD;骨组织计量(Micro-CT)方法测定骨微结构。结果:去卵巢术后各组大鼠BMD下降明显,松质骨、皮质骨骨量相继减少,皮质骨变薄;骨组织体积及骨小梁体积分数、厚度、宽度、数目及模块因素下降明显,分离度升高明显;适当剂量蒙药蓝刺头可提高大鼠骨组织体积、骨小梁体积分数、厚度、宽度、数目及模块因素,降低骨小梁分离度;有明显抗PMOP作用。结论:蒙药蓝刺头具有抗PMOP模型大鼠骨质疏松作用,能降低PMOP模型大鼠骨吸收,可为蒙医药补暖补精-清镇赫依-固骨质壮骨治法防治PMOP的科学研究提供实验依据。 Objective:To provide experimental reference for the prevention and treatment for postmenopausal osteoporosis(PMOP)based on the classical theory of mongolian medicine,and provide the thinking for mongolian medicine modernization.Methods:All 49 healthy rats aged four months were randomized into sham operation group,the model group,Yinyanghuo group,coenzyme Q10 group,high,moderate and low dose groups of mongolian medicine Lancitou(echinops sphaerocephalus).PMOP models were established by castrating,vaginal cornification test was used to testify whether modeling succeeded.The model group and sham operation group were drenched with normal saline 90 days after the surgery,other groups with the corresponding herbs.Right intact femur was collected,fixed and conserved with 4%paraformldehyde solution after the rats were sacrificed.To detect bone mineral density(BMD)of right femur,including femoral proximal BMD and total BMD of the femur;bone tissue metrology was used to measure bone microstructure.Results:After ovariectomized,BMD reduced in different groups,bone mass reduced successively in spongy bone and cortical bone,cortical bone became thinner;after ovariectomized,bone tissue volume and bone trabuculae volume fraction,thickness,width,number and module in different groups lowered notably,resolution rose notably;adequate dose of mongolian medicine Lancitou could raise bone volume,bone volume/tissue volume(BV/TV),thickness,width,number and patter factor,and decrease trabuculae spacing;and it possessed remarkable anti-PMOP effects.Conclusion:Mongolian medicine Lancitou could fight against osteoporosis of PMOP rats remarkably,apparently decrease bone resorption of PMOP rat model,and it could provide experimental reference for scientific study on the prevention and treatment for PMOP by classical treatment of mongolian medicine.
