基于网络药理学研究淫羊藿-女贞子药对抗骨质疏松作用机制 被引量:3

Mechanism Analysis of Herba Epimedii/Fructus LigustriLucidi against Osteoprosis Using Network Pharmacology

摘要 目的利用网络药理学探讨淫羊藿-女贞子药对抗骨质疏松的作用机制。方法采用中药系统药理学数据库(TCMSP)筛选淫羊藿-女贞子药对活性成分,预测活性成分对应的靶蛋白和相互作用蛋白,与来源于CTD和TTD数据库的骨质疏松靶蛋白和相互作用蛋白进行交集获取核心靶蛋白,利用cytoscape构建活性成分-靶蛋白网络,成分-靶点-疾病交互网络,利用DAVID数据库富集核心靶蛋白的相关信号通路。结果检索淫羊藿-女贞子药对活性成分19个,筛选53个核心靶蛋白,度值最高的核心靶蛋白为雌激素受体1(ESR1),获取32条相关信号通路。结论淫羊藿-女贞子药对可通过调节多个与成骨细胞、破骨细胞功能相关的信号途径而起到抗骨质疏松作用,靶蛋白ESR1及其相关信号通路可能较为重要。 Objective To investigate the anti 一 osteoprosis mechanism of herba epimedii/fructus ligustri lucidi by using network pharmacology. Methods Traditional Chinese medicine systems pharmacology (TCMSP) database was used to search for the active components of herba epimedii/fructus ligustri lucidi and to predict the target proteins and interaction proteins of active components. TCMSP database was combined with CTD and TTD database to obtain the core target proteins and the interaction proteins related to osteoporosis. DAVID database was used to enrich the related signaling pathways of core target proteins. Results 249 chemical components and 19 active components were screened. The component target network and component target disease network were constructed by using Cytoscape. 53 core target proteins were screened and the core target protein ESRI showed the highest degree value. 32 signaling pathways were obtained finally. Conclusion The therapeutic effect of herba epimedii/fructus ligustri lucidi on osteoprosis is regulated by signaling pathways involved in the functions of osteoblast and osteoclast. ESRI protein and its signaling pathways may play an important role.
