子宫内膜样腺癌组织hCLOCK蛋白表达临床意义

hCLOCK protein expression of endometrioid adenocarcinoma and its clinical significance

摘要 目的人类生物钟(human circadian clock,hCLOCK)蛋白作为节律调节蛋白家族的核心成员,在细胞的转录调控中发挥重要作用,研究表明其与肿瘤细胞的增殖和转移相关。本研究分析了hCLOCK蛋白在子宫内膜癌组织中的表达水平及临床意义。方法收集2015-11-01-2018-11-30滨州医学院附属医院妇科手术切除的子宫内膜癌及癌旁组织共97对,采用免疫组化方法检测hCLOCK蛋白在肿瘤及癌旁组织中的表达水平,采用χ^2检验和Spearman秩次相关分析其表达与临床病理的关系。结果 hCLOCK蛋白在子宫内膜癌组织中阳性表达率为63.92%(62/97),在癌旁组织中为42.27%(41/97),两者比较差异有统计学意义,χ^2=9.128,P=0.003。相关分析显示,hCLOCK蛋白表达水平与患者的淋巴结转移(rs=0.218,P=0.032)、FIGO分期(rs=0.331,P=0.001)及肌层浸润(rs=-0.213,P=0.036)有低度统计学意义的相关,而与年龄(rs=-0.042,P=0.679)、组织学分化(rs=-0.145,P=0.158)、ER(rs=-0.172,P=0.092)、PR(rs=-0.186,P=0.068)无统计学意义的相关。结论 hCLOCK蛋白在子宫内膜癌组织中呈现高表达,与肿瘤分期、淋巴结转移和肌层浸润有关。 OBJECTIVE As a core member of the rhythm regulation protein family,human circadian clock(hCLOCK)protein plays an important role in the regulation of cell transcription.Studies have shown that it is related to tumor cell proliferation and metastasis.This study analyzed the expression level and clinical significance of hCLOCK protein in endometrial cancer.METHODS A total of 97 pairs of endometrial cancer tissues and adjacent tissues that were surgically excised in department of gynecology of Binzhou medical university hospital from November 1,2015 to November 30,2018 were collected.The expression level of hCLOCK protein in tumor tissues and adjacent tissues was detected by immunohistochemistry.Chi-square test and Spearman rank correlation analysis were used to analyze the relationship between expression and clinicopathology.RESULTS The positive expression rate of hCLOCK protein in endometrial cancer tissues was 63.92%(62/97),and the positive expression rate in adjacent tissues was 42.27%(41/97).Comparison of positive rates between the two,the difference was statistically significant(χ^2=9.128,P=0.003).Correlation analysis showed that the hCLOCK protein expression level was low correlated with lymph node metastasis(rs=0.218,P=0.032),FIGO stage(rs=0.331,P=0.001),and muscle infiltration(rs=-0.213,P=0.036),but not with age(rs=-0.042,P=0.679),histological differentiation(rs=-0.145,P=0.158),ER(rs=-0.172,P=0.092)or PR(rs=-0.186,P=0.068).CONCLUSION hCLOCK protein is highly expressed in endometrial cancer tissues and is related to FIGO stage,lymph node metastasis and muscle infiltration.
