摘要 目的:探讨外源性高迁移率蛋白(High Mobility Group Box 1,HMGB1)对绒毛外滋养细胞(HTR8/SVneo细胞)侵袭和迁移能力的影响。方法:①确定HMGB1对绒毛外滋养细胞侵袭性的影响HTR8/SVneo细胞随机分为正常对照组及50μg/L、100μg/L和200μg/L外源性HMGB1刺激组,分别培养6 h、12 h、24 h、48 h,收集不同时间点不同刺激浓度的细胞,采用Transwell体外侵袭实验检测HTR8/SVneo细胞的侵袭性。②确定HMGB1对细胞增殖的影响:细胞随机分为正常对照及50μg/L、100μg/L和200μg/L外源性HMGB1刺激组.分别培养6 h、12 h、24 h、48 h,收集不同时间点不同刺激浓度的细胞,以CCK8方法检测细胞的增殖水平。结果:①HMGB1对绒毛外滋养细胞侵袭性的影响:收集不同时间点、不同刺激浓度的细胞,统计不同时间点、不同刺激浓度的细胞数量,不同时间之间差异有统计学意义(F=204.076,P<0.001);时间与刺激浓度之间存在交互效应(F=16.158,P<0.001);②HMGB1对细胞增殖的影响:不同时间因素之间有统计学意义(F=116.853,P<0.001);时间与处理因素之间不存在交互效应(F=1.933,P>0.05)。结论:外源性HMGB1可以增强绒毛外滋养细胞株HTR8/SVneo的侵袭能力和迁移能力。 Objective:To investigate the effect of HMGB1 on the invasion and migration of extracellular trophoblast and its related mechanism.Methods:To determine the effect of HMGB1 on the invasiveness of extracellular trophoblast cells,HTR8/SVneo cell was randomly divided into the normal control group and 50μg/L,100μg/L and 200μg/L exogenous HMGB1 groups,cultured for 6 hours,12 hours,24 hours and 48 hours,respectively.The invasiveness of HTR8/SVneo cells was detected by Transwell invasion assay in vitro.To determine the effect of HMGB1 on cell proliferation:the cells were randomly divided into normal control and 50μg/L 100μg/L and 200μg/L exogenous HMGB1 stimulation groups,cultured for 6 hours,12 hours,24 hours and 48 hours,respectively.The level of cell proliferation was detected by CCK8 method.Results:To determine the effect of HMGB1 on the invasiveness of extracellular trophoblastic cells,collecting cells at different time points and different concentrations of stimulation,and counting the number of cells at different time points and different concentrations of stimulation,there was a significant difference between different time points(F=204.076,P<0.001);There was interaction between time and stimulation concentration(F=16.158,P<0.001).The effect of HMGB1 on cell proliferation was determined,there was statistical difference among different time factors(F=116.853,P<0.001),and there was no interaction between time and treatment factors(F=1.933,P>0.05).Conculusion:Exogenous HMGB1 enhanced the invasion and migration ability of HTR8/SVneo.
