Preliminary study of HMGB1-RAGE signaling pathway in the pathogenesis of preeclampsia

摘要 目的探讨HMGB1-RAGE信号通路相关因子HMGB1、RAGE、NF-κBp65在子痫前期发病机制中的作用。方法选择2017年8月至2018年1月在郑州大学第一附属医院产科分娩的子痫前期孕妇60例,按病情轻重分为子痫前期组(轻度组30例),重度子痫前期组(重度组30例),另选择同期健康孕妇30例为正常对照组。检测各组血清、胎盘中HMGB1、RAGE、NF-κBp65的表达。结果 HMGB1、RAGE、NF-κBp65均表达于绒毛滋养细胞及绒毛外滋养细胞,HMGB1主要表达于胎盘组织滋养细胞的细胞核,RAGE、NF-κBp65主要表达于细胞质和细胞膜,轻度组及重度组的阳性表达率均高于对照组,差别有统计学意义(P <0.05)。轻度组、重度组胎盘组织及外周血中HMGB1 mRNA、RAGE mRNA及相应蛋白的表达均高于对照组(P <0.05)。血清中HMGB1、RAGE蛋白水平与子痫前期孕妇收缩压呈正相关(r=0.389和r=0.550)。结论子痫前期及重度子痫前期患者胎盘及血清中HMGB1、RAGE、NF-κBp65蛋白表达升高,HMGB1-RAGE炎症反应通路可能参与子痫前期的发生与发展。 Objective To investigate the role of relevant factors of HMGB1、RAGE、NF-κBp65 in HMGB1-RAGE signaling pathway in the pathogenesis of preeclampsia.Methods Sixty patients with preeclampsia were recruited from August 2017 to January 2018 in the First Affiliated Hospital of Zhengzhou University.Thirty cases were defined as the mild preeclampsia group and thirty cases were defined as the severe preeclampsia group.The other thirty healthy pregnant women were also recruited.The expressions of HMGB1,RAGE and NF-κBp65 were detected in the serum and placenta of each group.Results HMGB1,RAGE and NF-κBp65 was observed in the cytoplasm of cytotrophoblast in the three groups.HMGB1 was mainly expressed in the nucleus of trophoblast cells in placental tissue.RAGE and NF-κBp65 were mainly expressed in the cytoplasm and cell membrane of trophoblast cells.The positive rates of placenta in the mild and severe preeclampsia groups were signif icantly higher than that in the control group(P <0.05).The expressions of HMGB1 mRNA,RAGE mRNA and protein in placental tissues of mild and severe groups were all higher than those of the control group(P <0.05).The expression of HMGB1 and RAGE in peripheral blood of the mild preeclampsia and severe preeclampsia groups was higher than that of the control group(P <0.05),and the level of HMGB1 and RAGE in serum were positively correlated with the systolic blood pressure of preeclampsia pregnant women(r=0.389 and r=0.550).Conclusions The increased expression of HMGB1、RAGE and NF-κBp65 were found in the blood and placentas in the preeclampsia.HMGB1-RAGE signaling pathway may plays an important role in promoting the occurence and development of preeclampsia.
