益气除痰方抑制NK/T细胞淋巴瘤及PI3K/AKT/mTOR信号通路的研究 被引量:3

Inhibitory Effects of Yiqichutan Decoction on NK/T Cell Lymphoma and PI3K/AKT/mTOR Signaling Pathway

摘要 目的:研究益气除痰方对NK/T细胞淋巴瘤生长及PI3K/AKT/m TOR信号通路激活的抑制作用。方法:培养人NK/T淋巴瘤细胞的细胞株SNK-6,制备益气除痰方含药血清,用含有35%空白血清、15%、25%、35%益气除痰方63 g/kg含药血清或35%益气除痰方63 g/kg含药血清+25 ng/m L胰岛素样生长因子-1的培养基处理,检测细胞增殖活力、细胞周期、PI3K/AKT/m TOR通路分子表达量;制备SNK-6细胞的移植瘤小鼠,以27. 3 g/kg、54. 6 g/kg益气除痰方灌胃干预后检测移植瘤质量及PI3K/AKT/m TOR通路增殖活力表达。结果:与空白血清组比较,15%、25%、35%益气除痰方63 g/kg含药血清组的蛋白、S期比例明显降低及p-PI3K、p-AKT、p-mTOR的蛋白表达量明显下调,G0/G1期比例明显增加(P <0. 05或P <0. 01);与35%益气除痰方63 g/kg含药血清组比较,35%益气除痰方含药血清+25 ng/m L胰岛素样生长因子-1组的增殖活力值、S期比例明显增加及p-PI3K、p-AKT、pm TOR的蛋白表达量明显上调,G0/G1期比例明显减少(P <0. 05或P <0. 01)。与模型对照组比较,27. 3 g/kg、54. 6 g/kg益气除痰方组移植瘤小鼠的移植瘤质量明显减轻,移植瘤中p-PI3K、p-AKT、p-mTOR的蛋白表达量明显下调(P <0. 05或P <0. 01)。结论:益气除痰方能够抑制NK/T细胞淋巴瘤生长且该抑制作用与抑制PI3K/AKT/m TOR信号通路有关。 Objective: To study the inhibitory effect of Yiqichutan decoction on the growth of NK/T cell lymphoma and the activation of PI3 K/AKT/m TOR signaling pathway. Methods: SNK-6 cell line of human NK/T cell lymphoma cells was cultured and the serum containing Yiqichutan Decoction was prepared. The cells were treated with medium containing 35 % blank serum,63 g/kg drug-containing serum( 15%,25% and 35 %) or 35 % drug-containing serum + 25 ng/m L insulin-like growth factor-1. Then the cell proliferation activity,cell cycle,molecule expression of PI3 K/AKT/m TOR signaling pathway were detected. SNK-6 cell transplanted tumor mice were prepared and 27. 3 g/kg or 54. 6 g/kg Yiqichutan decoctions were intragastrically administered. The mass of transplanted tumor and the molecule expression level of PI3 K/AKT/m TOR pathway were detected. Results: Compared with the control group,the protein expression,S phase ratio and the expressions of p-PI3 K,p-AKT and p-m TOR in 15%,25% and 35% drug-containing serum( 63 g/kg) groups was significantly decreased,while the G0/G1 phase ratio was significantly increased. Compared with 35% drug-containing serum group,the proliferation activity,S phase ratio and the protein expressions of p-PI3 K,p-AKT and p-m TOR in 35% drug-containing serum + 25 ng/m L insulin-like growth factor-1 group were significantly increased,while the G0/G1 phase ratio was significantly decreased. Compared with the control group,the mass of transplanted tumors in 27. 3 g/kg or 54. 6 g/kg Yiqichutan decoction groups were significantly reduced,the protein expressions of p-PI3 K,p-AKT and p-m TOR transplanted tumors were significantly decreased. Conclusion: Yiqichutan Decoction could inhibit the growth of NK/T cell lymphoma,its mechanisim may be related to the inhibition of PI3 K/AKT/m TOR signaling pathway.
