摘要
目的探讨白藜芦醇(resveratrol,RES)在伯基特淋巴瘤细胞株中,对IL-6/JAK2/STAT3信号通路的抑制作用。方法体外培养伯基特淋巴瘤Daudi细胞株,CCK-8检测不同浓度RES对Daudi细胞的活性变化,细胞组织免疫化学(SP法)检测Daudi细胞株中信号转导分子p-JAK2、p-STAT3的表达变化;免疫印迹(Western blotting)检测p-JAK2、p-STAT3以及凋亡相关蛋白Bcl-2、Survivin、Bax的表达变化。结果 RES处理组与正常对照组比较,Daudi细胞的活性随着RES浓度和作用时间的增加而降低;RES预处理24 h,IL6(interleukin 6)处理30 min,与单独用IL-6处理比较,p-JAK2、p-STAT3以及抗凋亡因子Bcl-2、Survivin的表达明显降低,而促凋亡因子Bax的表达明显升高(P<0.05)。结论在伯基特淋巴瘤中,RES可能通过下调由IL6引起的p-JAK2、p-STAT3的表达,提高促凋亡因子的表达,降低抗凋亡因子的表达,促进肿瘤细胞的凋亡,发挥抗肿瘤的作用。
Objective To investigate the regulative mechanism of resveratrol(RES) on JAK2/STAT3 signal pathway in burkitt's lymphoma Daudi cells. Methods Human burkitt's lymphoma Daudi cells were cultured in vitro. The cells were treated by different concentration of RES for different time, and the cell proliferation rate was evaluated by CCK-8 assay. The variation of expression levels of signal transduction factor p-JAK2 and p-STAT3 in Daudi cell line was analyzed by immunocytochemistry. Expression levels of p-JAK2 and p-STAT3 and apoptosis-related protein Survivin,Bcl-2 and Bax were measured by Western blotting method. Results Daudi cell proliferation rate decreased with the increase of RES concentration and action time in RES group. After RES pretreatment for 24 h and treatment with IL-6 for 30 min,the expression levels of p-JAK2,p-STAT3,Bcl-2 and Survivin were much decreased,while Bax expression level was increased obviously compared to the group treated by IL-6 only(P〈0.05). Conclusion In the burkitt's lymphoma,RES promotes the apoptosis of Daudi cell line probably through down-regulating IL-6 to resist the JAK2/STAT3 signaling pathway, thus to promote the expression of apoptosis-associated factors and inhibit anti-apoptosis factors.
