摘要
目的建立气相色谱检测法测定南瓜中肌醇的方法。方法样品粉碎匀浆后,经水+无水乙醇(15:35,V:V)超声提取20 min,涡旋振荡25 min浓缩烘干,复溶后进行衍生化。用氢火焰离子化检测器检测,外标法定量。结果肌醇衍生物在10~200 mg/L范围内线性关系良好,相关系数r 2为0.99996。检出限与定量限分别为1.0、3.0 mg/kg,回收率为88.5%~100.3%,相对标准偏差小于5%,使用上述方法对23批次南瓜样品进行肌醇含量检测,结果为784~1792 mg/kg。结论该方法高效,便捷,准确度高,适用于南瓜等各类蔬菜水果中肌醇的检测。
Objective To establish a method for the determination of inositol in pumpkin by gas chromatography.Methods After the sample was crushed and homogenized,it was extracted with water and absolute ethanol(15:35,V:V)ultrasonic for 20 min,vortexed for 25 min,concentrated and dried,and derivatized after redissolving.It is detected by hydrogen flame ionization detector and quantified by external standard method.Result The inositol derivative had a good linear relationship in the range of 10–200 mg/L,and the correlation coefficient r2 was 0.99996.The limit of detection and the limit of quantification were 1.0 mg/kg and 3.0 mg/kg,respectively.The recovery rates were 88.5%–100.3%,and the relative standard deviation was less than 5%.The inositol content of 23 batches of pumpkin samples was tested and the results were 784–1792 mg/kg.Conclusion This method is efficient,convenient,and accurate for determination of inositol in pumpkin as well as other fruits and vegetables.
