灰树花杂交菌株鲁灰86的选育 被引量:2

摘要 以灰树花(Grifola frondosa)野生菌株梯灰1号和栽培菌株庆灰151为亲本,各取10个孢子单核体单单杂交,采用光学显微镜观察是否存在锁状联合结构及通过与亲本的拮抗实验最终获得65个杂交子。选取23个菌丝生长速度较快的杂交子进行小试初筛栽培实验,并采用ISSR和SRAP分子标记对筛选出的优良杂交子构建系统发育树。结果表明:鲁灰86子实体颜色较深、朵型较大、菌管较短,产量分别比亲本梯灰1号和庆灰151提高了74.47%和20.53%,生物学转化率大于50%,且子实体中多糖含量较高;鲁灰86与两亲本间存在明显的遗传差异,是具有推广潜力的优质高产菌株。 Using Grifola frondosa wild-type strain Tihui No. 1 and cultivar Qinghui 151 as the parent strains, mon-mon hybridization was conducted with 10 spore monokaryons of each strain to cross with the other strain. Then 65 real hybrids were obtained by clamp connection observation and antagonism reaction with their parents. Twenty-three hybrids with high mycelial growth rate were selected for preliminary screening cultivation. Seven of these selected hybrids were further subjected to phylogenetic analysis using both ISSR and SRAP molecular markers. The results showed that one of the hybrids designated Luhui 86 had big fruiting body clusters, dark fruiting bodies with high content of polysaccharides and short tubules. Compared with Tihui No. 1 and Qinghui 151, the average yield of Luhui 86 was increased by 74.47% and 20.53%, respectively. The biological efficiency of Luhui 86 was over 50%. There were significant genetic differences between Luhui 86 and its parents. With high quality and yield, Luhui 86 has a high potential to be used in cultivation.
