摘要 目的探讨TGF-β1/Smad信号通路在人参养荣汤抑制血管紧张素Ⅱ诱导的心肌细胞病理性肥大中的作用。方法体外培养H9C2心肌细胞,采用AngⅡ诱导心肌细胞肥大,采用MTT实验观察人参养荣汤对病理性肥大心肌细胞活力的影响。采用qPCR检测心肌细胞中ANF、α-SMA mRNA的表达水平,Western blot检测心肌细胞中TGF-β1/Smad信号通路表达水平。结果AngⅡ处理24 h后能够诱导H9C2心肌细胞肥大,增强细胞活力,增加细胞内ANF、α-SMA mRNA的表达水平,以及上调TGF-β1/Smad信号通路。人参养荣汤能够抑制AngⅡ诱导的心肌细胞肥大,下调心肌细胞中ANF、α-SMA mRNA的表达水平,以及抑制TGF-β1/Smad信号通路。结论人参养荣汤能够抑制AngⅡ诱导的心肌细胞肥大,TGF-β1/Smad信号通路可能是人参养荣汤发挥抗心肌肥大的作用靶点。 Objective To investigate the role of TGF-β1/Smad signaling pathway in the inhibition of angiotensinⅡ-induced cardiomyocyte hypertrophy by Renshen Yangrong Decoction.Methods H9C2 cardiomyocytes were cultured in vitro,and cardiomyocyte hypertrophy was induced by AngⅡ.The effect of Renshen Yangrong Decoction on the activity of pathological hypertrophic cardiomyocytes was observed by MTT assay.The expression levels of ANF andα-SMA m RNA in cardiomyocytes were detected by q PCR.The expression of TGF-β1/Smad signaling pathway in cardiomyocytes was detected by Western blot.Results After 24 hours of AngⅡtreatment,H9C2 cardiomyocyte hypertrophy was induced,cell viability was enhanced,ANF andα-SMA m RNA expression levels were increased,and TGF-β1/Smad signaling pathway was up-regulated.RenshenYangrong Decoction can inhibit AngⅡ-induced cardiomyocyte hypertrophy,down-regulate the expression of ANF andα-SMA mRNA in cardiomyocytes,and inhibit TGF-β1/Smad signaling pathway.Conclusion Renshen Yangrong Decoction can inhibit AngⅡ-induced cardiomyocyte hypertrophy.TGF-β1/Smad signaling pathway may be the target of anti-myocardial hypertrophy by Renshen Yangrong Decoction.
