miR-451对膀胱癌细胞迁移、侵袭及E-cadherin和Vimentin基因表达的调控作用 被引量:4

Regulation effect of miR-451 on migration,invasion and e-cadherin and vimentin gene expression in bladder cancer cells

摘要 目的研究miR-451对膀胱癌细胞迁移、侵袭及E-钙黏蛋白(E-cadherin)和波形蛋白(Vimentin)基因表达的调控作用。方法培养人膀胱癌细胞株T24、5637、SW790并采用荧光定量PCR检测miR-451的表达量;T24细胞随机分为不转染模拟物的对照组、转染NC模拟物的NC组、转染miR-451模拟物的miR-451组,采用荧光定量PCR检测miR-451的相对表达量,采用Western blot检测E-cadherin和Vimentin的相对表达量,采用Transwell检测细胞的迁移、侵袭能力。结果T24细胞中miR-451的相对表达量均低于5637细胞、SW790细胞;在T24细胞中,miR-451组的miR-451相对表达量、E-cadherin相对表达量明显高于对照组、NC组,迁移能力、侵袭能力均明显弱于对照组、NC组,Vimentin相对表达量明显低于对照组、NC组。结论miR-451能够抑制膀胱癌细胞的迁移、侵袭且该抑制作用与增加E-cadherin表达、减少Vimentin表达有关。 Objective To study the regulation effect of miR-451 on the migration,invasion and expression of E-cadherin and Vimentin genes in bladder cancer cells.Methods Human bladder cancer cell lines T24,5637,SW790 were cultured and the expression of miR-451 was detected by fluorescent quantitative PCR.T24 cells were randomly divided into control group without transfection,NC group transfected with NC mimic and miR-451 group transfected with miR-451 mimic.The relative expression of miR-451 was detected by fluorescent quantitative PCR and the relative expression of E-cadherin and Vimentin were detected by Western blot,the migration and invasion ability of cells were detected by transwell.Results The relative expression of miR-451 in T24 cells was lower than those in 5637 cells and SW790 cells.In T24 cells,the relative expression of miR-451 and E-cadherin in the miR-451 group were significantly higher than those in the control group and NC group,and the migration and invasion ability were significantly weaker than those in the control group and NC group,while the relative expression of Vimentin was significantly lower than that in the control group and NC group.Conclusions miR-451 can inhibit the migration and invasion of bladder cancer cells,and the inhibition effect is related to the increase of E-cadherin expression and the decrease of Vimentin expression.
