摘要
目的:研究补阳还五汤对脑出血大鼠模型基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制剂-1(TIMP-1)、谷氨酸(Glu)及PI3K、AKT表达的作用。方法:选取90只SD大鼠,以随机抽签法分成模型组、正常对照组及补阳还五汤组。模型组与补阳还五汤组建立脑出血模型,造模后第2 d。补阳还五汤组行补阳还五汤灌胃,模型组行生理盐水灌胃,给药14 d。比较3组MMP-9、TIMP-1、Glu,脑组织PI3K、AKT以及Bcl-2、BAX蛋白表达。结果:补阳还五汤组、模型组的MMP-9、TIMP-1、Glu均高于正常对照组,且补阳还五汤组MMP-9、Glu低于模型组,而TIMP-1高于模型组(均P<0.05);补阳还五汤组、模型组脑组织PI3K、AKT蛋白表达较正常对照组更高,补阳还五汤组脑组织PI3K、AKT蛋白表达较模型组更高(均P<0.05);补阳还五汤组、模型组脑组织Bcl-2、BAX蛋白表达均高于模型组,补阳还五汤组脑组织Bcl-2、BAX蛋白表达较模型组更高(均P<0.05)。结论:补阳还五汤可改善脑出血大鼠MMP-9、TIMP-1、Glu,可由激活PI3K/AKT信号转导通路,调控Bcl-2、BAX表达,继而发挥保护脑组织作用。
Objective:To investigate and analyze the effects of Buyanghuanwu Decoction on the expression of matrix metalloproteinase-9(MMP-9),matrix metalloproteinase inhibitor-1(TIMP-1),glutamate(Glu),PI3 K and AKT in rats with cerebral hemorrhage.Methods:A total of 90 SD rats were selected and divided into a model group,a normal control group and Buyang Huanwu Decoction group by random lottery.Rats in the model group and the Buyang Huanwu Decoction group were given the cerebral hemorrhage model.Starting from 2 d after the model,the Buyang Huanwu Decoction group was given the gavage treatment of Buyang Huanwu Decoction,while the model group was given normal saline and continuous administration for 14 d.The expression levels of MMP-9,TIMP-1,Glu,PI3 K,and AKT,Bcl-2,BAX in brain tissues of the 3 groups of rats were compared.Results:The levels of MMP-9,TIMP-1 and Glu in the Buyang Huanwu Decoction group and the model group were higher than those in the normal control group,and the levels of MMP-9 and Glu in the Buyang Huanwu Decoction group were lower than those in the model group while the TIMP-1 was higher than those in the model group(all P<0.05).The expression levels of PI3 K and AKT in the brain tissues of rats in the Buyang Huanwu Decoction group and the model group were higher than those in the normal control group.PI3 K、AKT protein expression levels in brain tissues of rats in Buyang Huanwu Decoction were higher than those in the model group(all P<0.05).Bcl-2 and BAX protein expression levels in brain tissues of rats in Buyang Huanwu Decoction group were higher than those in model group(both P<0.05).Conclusion:Buyang Huanwu Decoction can significantly improve the levels of MMP-9,TIMP-1 and Glu in rats with cerebral hemorrhage,which may further regulate the expression of Bcl-2 and BAX proteins by activating the PI3 K/AKT signal transduction pathway,thereby playing a role in protecting brain tissue.
