摘要
目的:研究口服阿莫西林和头孢呋辛酯分别对蒲地蓝消炎口服液中黄芩苷血药浓度的影响。方法:用液相色谱质谱联用(HPLC-MS-MS)法测定阿莫西林和头孢呋辛酯分别与蒲地蓝消炎口服液合并给药组与蒲地蓝消炎口服液单独给药组大鼠的血药浓度,比较3者的药动学参数。色谱柱为Phenomenex Luna C8(2.0 mm×50 mm,3μm),流动相为甲醇-0.05%甲酸,1%异丙醇水溶液,采用梯度洗脱,体积流量为200μl·min^-1。质谱采用电喷雾离子源,多反应监测方式进行负离子扫描。结果:阿莫西林和头孢呋辛酯与蒲地蓝消炎口服液合用后均能显著降低蒲地蓝消炎口服液中黄芩苷的达峰浓度Cmax及曲线下面积AUC0-36h(P<0.05)。结论:联用阿莫西林或头孢呋辛酯可影响蒲地蓝消炎口服液中黄芩苷的血药浓度。
Objective: To study the influence of oral amoxicillin and cefuroxime axetil on baicalin level in rat plasma. Methods:Rats were divided into three groups. The first group was treated with amoxicillin and baicalin in combination,the second group was treated with cefuroxime axetil and baicalin in combination,and the third group was treated with baicalin. The baicalin concentration in the three groups was detected by HPLC-MS-MS. The pharmacokinetic parameters were calculated by statistical analysis and compared.The determination was performed on a Phenomenex Luna C8( 2.0 mm×50 mm,3 μm) with gradient program. The mobile phases consisted of methanol and water( containing 0.05% formic acid and 1% isopropanol) at a flow rate of 200μl·min^-1. Electrospray ionization( ESI) and multiple reaction monitoring( MRM) were used in a positive ion mode. Results: Amoxicillin and cefuroxime axetil combined with Pudilan antiphlogistic oral liquid significantly reduced the Cmaxand AUC0-36 hof baicalin in Pudilan antiphlogistic oral liquid( P<0.05). Conclusion: Amoxicillin and cefuroxime axetil administrated orally can influence the baicalin concentration in rat’s plasma significantly.
