摘要
背景动物实验研究表明,左心室心肌组织骨形成蛋白和激活素的跨膜抑制剂(BAMBI)表达上调可对抗转化生长因子β1(TGF-β1)的致心肌纤维化作用,而心肌纤维化又与室性心律失常(VA)密切相关,因此推测BAMBI和TGF-β1可能参与了高血压患者VA的发生发展过程。目的分析自发性高血压大鼠左心室心肌组织BAMBI、TGF-β1表达情况及其与VA易感性的关系。方法2018年7月-2019年7月,选取6只14周龄雄性Wistar大鼠作为对照组,6只与Wistar大鼠同源的雄性自发性高血压大鼠作为实验组;两组大鼠均饲养至22周龄。比较两组大鼠22周龄时收缩压、心率、心脏彩超检查结果〔包括左心室舒张末期内径(LVEDD)、左心室收缩末期内径(LVESD)、左心室射血分数(LVEF)及左心室短轴缩短率(LVFS)〕、VA诱发成功率,左心室心肌组织BAMBI、TGF-β1 mRNA及其相对表达量。结果两组大鼠22周龄时心率和LVEDD比较,差异无统计学意义(P>0.05);实验组大鼠22周龄时收缩压高于对照组,LVESD大于对照组,LVEF和LVFS低于对照组(P<0.05)。实验组大鼠22周龄时VA诱发成功率高于对照组(P<0.05)。实验组大鼠22周龄时左心室心肌组织BAMBI和TGF-β1 mRNA及其相对表达量均高于对照组(P<0.05)。结论22周龄自发性高血压大鼠左心室心肌组织中BAMBI和TGF-β1表达上调可导致心肌纤维化加重、左心室重构,进而增加VA易感性。
Background Animal studies showed that,up-regulated expression of BAMBI in left ventricular myocardium can counteract the myocardial fibrosis caused by TGF-β1,however myocardial fibrosis is closely related to ventricular arrhythmia(VA),therefore it is being speculated that BAMBI and TGF-β1 may be involved in the occurrence and development of VA in patients with hypertension.Objective To analyze the expressions of BAMBI and TGF-β1 in left ventricular myocardial tissue and their relations to susceptibility of VA in spontaneously hypertensive rats(SHR).Methods From July 2018 to July 2019,a total of six 14 week-old male Wistar rats were selected as control group,meanwhile six 14 week-old homologous male SHR rats were selected as experiment group;rats in the two groups were raised to 22 weekold.SBP,heart rate,color Doppler echocardiography examination results(including LVEDD,LVESD,LVEF and LVFS),success rate of induction of VA,BAMBI and TGF-β1 mRNA and their relative expression quantity in left ventricular myocardial tissue were compared between the two groups at 22 weeks old.Results There was no statistically significant difference in heart rate or LVEDD between the two groups at 22 weeks old(P>0.05);SBP in experiment group was statistically significantly higher than that in control group,LVESD in experiment group was statistically significantly greater than that in control group,while LVEF and LVFS in experiment group were statistically significantly lower than those in control group(P<0.05).Success rate of induction of VA in experiment group was statistically significantly higher than that in control group at 22 weeks old(P<0.05).BAMBI and TGF-β1 mRNA and their relative expression quantity in left ventricular myocardial tissue in experiment group were statistically significantly higher than those in control group at 22 weeks old,respectively(P<0.05).Conclusion Expressions of BAMBI and TGF-β1 in left ventricular myocardial tissue are up-regulated in 22 week-old SHR,which may lead to the aggravation of myocardia
