miR-19a调控上皮-间质转化对结肠炎相关性结直肠癌的影响 被引量:4

摘要 目的探讨miR-19a调控的上皮-间质转化(EMT)对结肠炎相关性结直肠癌(UCRCC)的影响。方法40只雄性C57BL/6J小鼠随机分为模型组、AS-miR-19a组、阴性对照组和正常对照组,模型组、AS-miR-19a组、阴性对照组建立氧化偶氮甲烷(AOM)/葡聚糖硫酸钠(DSS)诱导小鼠UCRCC模型,AS-miR-19a组于第7周结肠灌注miR-19a反义寡核苷酸腺病毒载体。记录小鼠体质量和存活情况,第11周处死小鼠,测量小鼠结肠长度、成瘤数量,检测结肠组织E-cadherin、N-cadherin蛋白表达。结果AS-miR-19a组小鼠存活率、体质量明显高于模型组和阴性对照组(均P<0.05)。AS-miR-19a组炎性反应较模型组和阴性对照组明显减轻(P<0.05)。与正常对照组比较,模型组和阴性对照组miR-19a表达明显增加,AS-miR-19a组miR-19a表达显著降低(均P<0.05)。AS-miR-19a组小鼠结肠长度、结肠成瘤数量明显低于模型组和阴性对照组(均P<0.05)。与模型组和阴性对照组比较,AS-miR-19a组E-cadherin蛋白表达显著增加,N-cadherin蛋白表达显著降低(均P<0.05)。结论miR-19a可能通过调控EMT介导UCRCC的发生。 Objective To investigate the effects of miR-19a mediated epithelial-mesenchymal transition(EMT)on ulcerative colitis-related colorectal cancer(UCRCC).Methods A total of 40 male C57BL/6J mice were randomly assigned to the model,AS-miR-19a,negative control and normal control groups.The UCRCC models induced by azoxymethane(AOM)in combination with dextran sulfate sodium(DSS)were established in the model,AS-miR-19a and negative control groups.At week 7,the AS-miR-19a group received colon perfusion of miR-19a antisense olignucleotides.The body weight and survival rate of mice were recorded.At week 11,the mice were sacrificed.The colon length and number of gross polyp/adenoma were recorded.The protein expressions of E-cadherin and N-cadherin were detected.Results Compared with the model and negative control groups,the AS-miR-19a group had significantly higher survival rate and body weight,weaker inflammatory reaction,longer colon,larger number of gross polyp/adenoma,increased expression of E-cadherin,and decreased expression of N-cadherin(all P<0.05).Compared with the normal control group,the model and negative control groups had notably higher expression of miR-19a,while the AS-miR-19a group showed an opposite trend(P<0.05).Conclusions miR-19a might contribute to ulcerative colitis-related colorectal cancer by regulating epithelial-mesenchymal transition.
