摘要
选取山东蓝思种业股份有限公司选育并初步认定的"非洲猪瘟抗性猪"蓝思白2号(LS-2)猪,通过实验室非洲猪瘟病毒(ASFV)强毒攻击试验,分析其抗感染应答特征。结果表明,LS-2猪对口服感染106.0TCID50ASFV SY18株具有明显抗性:6头LS-2猪中5头健活,仅在血液中出现间歇、短暂和低水平的病毒血症,并呈现较低程度的发热,随后恢复正常,从12 d开始出现针对病毒结构蛋白p54抗体,之后抗体水平持续升高;1头延迟发病死亡。而普通家猪于感染后3 d开始出现持续发热症状和病毒血症,10 d内全部死亡,期间p54抗体水平一直处于阴性或极低水平。抗性猪和普通家猪炎性因子的检测结果显示,主要抗炎因子如TGF-β2在LS-2抗性猪未感染前处于较低水平,感染后表现轻微(约1.5~2.0倍)升高,而在普通家猪未感染前处于高水平,感染后则表现为显著下降;在感染后6 d(潜伏后期),抗性猪主要促炎因子TNF-α出现轻微(约1.5~2.0倍)升高,而普通家猪则呈现数十至数百倍的显著增高。至试验结束时,LS-2猪临床表现正常,剖检未见内脏器官外观异常,实时PCR于肺、心、骨髓、胸腺等可检出ASFV核酸,但病毒分离结果呈阴性。组织切片染色显示,LS-2猪脾脏和肾脏结构正常,仅肺脏中肺泡呈现轻微融合、扩张。以上结果表明,和普通家猪相比,LS-2猪对ASFV人工感染具有明显抗性,该类型猪具有重大推广意义。
Lansibai-2(LS-2)pigs,which are bred by Shandong Landsee Genetics Co.,Ltd.,and initially thought to be African swine fever resistant,have been confirmed to possess the characteristics of anti-African swine fever virus(ASFV)infection through a trial in P3 laboratory.The results showed that LS-2 had obvious resistance to the oral challenge of virulent ASFV SY18 strain with dosage of 106.0 TCID50,out of 6 LS-2,5 pigs survived,with intermittent,transient and low level viremia,mild fever,and recovered.Antibodies against the viral structural protein p54 began to appear on 12 dpi and continued to rise until the observation end;1 out of 6 died after a delayed disease onset.While the common domestic pigs began to develop fever and viremia on the 3 rd day post infection,and all died within 10 d,during which the p54 antibody was negative or very low.The main anti-inflammatory factors,such as TGF-β2,were at a low level before infection in LS-2 pigs,and showed a slight increase of 1.5-2.0 times post infection,however,in common domestic pigs,it was at a high level before infection and decreased significantly post infection.While the main proinflammatory factor TNF-αin LS-2 pigs increased slightly(1.5-2.0 times)on 6 dpi,the common domestic pig showed significant increase of tens to hundreds of times than before infection.By the end of the trial,LS-2 pigs showed normal clinical performance,but ASFV DNA could be detected by real-time PCR,nonetheless virus isolation was negative.Tissue pathological sections showed that the spleen,heart and kidney of LS-2 were structurally normal,and the lungs become slightly thicken in alveolar diaphragm with a small amount of inflammatory cell infiltration.The above results showed that LS-2 had obvious resistance to ASFV infection compared with the common domestic pigs.This kind of pig is of significant importance for commercialization.
