摘要 目的探讨颗粒细胞中Lnc-ADAMTS1-1∶1的表达与多囊卵巢综合征(PCOS)患者脂代谢及胚胎质量的相关性。方法本研究纳入在郑州大学第一附属医院生殖医学中心行体外受精(IVF)治疗的35例PCOS组患者及24例排卵周期正常的对照组女性。使用实时PCR测得两组患者原代颗粒细胞中Lnc-ADAMTS1-1∶1的表达,并分析其与患者血清总胆固醇(TC)、低密度脂蛋白(LDL)和高密度脂蛋白(HDL)等临床指标以及卵母细胞成熟率和优胚率等IVF实验室指标的相关性。结果 PCOS组患者颗粒细胞中Lnc-ADAMTS1-1∶1的相对表达量(3.74±0.35)高于对照组(1.78±0.31),差异有统计学意义(P<0.05)。其表达水平与PCOS患者的血清TC、LDL呈显著负相关,与IVF优胚率呈显著正相关。结论 PCOS患者颗粒细胞中Lnc-ADAMTS1-1∶1的表达水平在一定程度上可反映患者的脂代谢状态,且其可能通过参与卵子发育的调控影响患者胚胎质量。 Objective To investigate the correlations between Lnc-ADAMTS1-1∶1 expressed in granulosa cells and lipid metabolism and embryo quality of patients with polycystic ovary syndrome(PCOS).Methods This study included 35 patients in the PCOS group and 24 women with normal ovulation cycles in the control group who underwent treatment of in vitro fertilization(IVF) in the reproductive medical center of the First Affiliated Hospital of Zhengzhou University. The expression level of Lnc-ADAMTS1-1∶1 in the primary granulosa cells of the two groups was measured by real-time PCR. The correlations between Lnc-ADAMTS1-1∶1 and the clinical indicators such as total cholesterol(TC), low density lipoprotein(LDL), high-density lipoprotein(HDL) and laboratory indicators such as oocyte maturation rate and good-embryo rate were analyzed.Results The relative expression level of Lnc-ADAMTS1-1∶1 in granulosa cells of PCOS group(3.74±0.35) was higher than that of control group(1.78±0.31), and the difference was statistically significant(P<0.05). It was significantly negatively correlated with TC and LDL, and it was positively correlated with the good-quality embryo rate of PCOS patients.Conclusion The expression level of Lnc-ADAMTS1-1∶1 in granulosa cells can reflect the lipid metabolism of PCOS patients, and its up-regulation may affect embryo quality by regulating oocyte development.
