不同产地当归药材9种成分的含量测定及主成分分析 被引量:26

Determination and Principal Component Analysis of Nine Components of Angelica sinensis from Different Origins

摘要 目的建立高效液相色谱(HPLC)法同时测定当归药材9种成分含量的方法,并评价不同产地样品的质量。方法采用Agilent TC-C18色谱柱(4.6 mm×150 mm,5μm);以0.2%甲酸(A)-乙腈(B)为流动相,梯度洗脱;流速:1.0 mL·min^-1;检测波长:280 nm。并用主成分分析法评价当归不同产地样品质量。结果所测当归药材中的9种成分(绿原酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、藁本内酯、欧当归内酯)分离度良好,10个不同产地的当归药材质量有显著差异。经主成分分析得3个主成分的累积贡献率达到88.043%。10个产地中岷县麻子川乡绿叶村一社的当归药材质量最优。结论所建立的HPLC法结合主成分分析法可综合评价当归药材的质量,且该方法准确简便、重复性好,可为当归药材的质量评价提供依据。 Objective The method for simultaneous determination of 9 components in Angelica sinensis by HPLC was established,and the quality of samples from different origins was evaluated. Methods The separation was performed on an Agilent TC-C18 column(4.6 mm×150 mm,5 μm) with gradient elution using 0.2% formic acid(A) and(B) acetonitrile as mobile phase at a flow rate of 1.0 mL·min^-1. The detection wavelength was 280 nm. Results The resolution of 9 components determined by HPLC was good,and the quality of Angelica sinensis from 10 different origins was significantly different. Three principal factors were obtained by principal component analysis,and the cumulative contribution rate of the three principal components reached 88.043%. The quality of Angelica sinensis from Mazichuan township Lüye village in Min County is the best among the 10 origins. Conclusion The method for simultaneous determination of 9 components of Angelica sinensis by HPLC combined with principal component analysis method can comprehensively evaluate the quality of Angelica sinensis;this method is accurate,simple and reproducible,which can provide reference for the quality evaluation of Angelica sinensis.
